Tongmai Dark Soup Study On The Mechanism Of Abnormal Blood Lipid Level In Rats With Coronary Heart Disease

Coronary heart disease (CHD) is one of the illnesses that endange human health,which is formed with dyslipidemia and other facters.With the long-term increased of blood serum cholesterin (TC),the rate of CHD happening adds.The level of blood serum low density lipoprotein cholesterin (LDL-C) inseasing is the main dangerous facter of CHD,while the high density lipoprotein cholesterin (HDL-C) is negatively related to the occurrence of CHD. Clinical practice often reduce TC and LDL, increase HDL to reduce the incidence of coronary heart disease risk.Traditiongal Chinese Medcine (TCM) consider that dyslipidemia belongs to phlegm and blood stasis category, to treat with the method of dissipating phlegm and removing blood stasis as the main principle, synchronic treating phlegm and blood stasis in the pectoral stuffiness pain and cardiodynia treatment has received recognition rifely for now.This research starts from the effect of Tongmaihua- zhuo decoction actted on blood lipid and myocardial ischemia in rats with CHD,to discuss the mechanism of synchronic treating phlegm and blood stasis in CHD treatment,to provide a new experiment gist for clinic application.Objective:To establish dyslipidemia and myocardial ischemia rats model ,and observe the therapeutic effects of Tongmaihuazhuo decoction with different dosages and atorvastatin on dyslipidemia and myocardial ischemia.Methods:1 Rats were divided into six groups stochasticly:normal group,model group,western medicine group, Tongmaihuazhuo decoction low,middle and high doseage group.Except for normal group rats,the other groups rats were lavaged into stomach with vitamin D3 70 10000IU/kg in three days and fed with high lipid feedstuff for 8 weeks to establish dyslipidemia rats model,the normal group were lavaged into stomach with the same volume brine and fed with normal feedstuff;8 weeks later ,all the rats were fed with normal feedstuff , before the getting material day , select 8 rats from each group everytime randomly ,except the normal group rats ,the other groups were injected intraperitoneally with pituitrin by 30U/kg to establish the myocardial ischemia model, the normal group were injected intraperitoneally with the same volume brine.2 The treatment groups rats were lavaged into stomach with atorvastatin (include atorvastatin 0.09mg/ml) and Tongmaihuazhuo decoction low,middle and high doseage (include Chinese herb doseage 0.6975g/ml,1.395g/ml,2.79g/ml) by 2ml/100g everyday separately , the normal group and model group rats were lavaged into stomach with the same volume brine , treat 28 days.3 On 7th,14th,28th days of treatment,to the rats which were injected intraperit- oneally with pituitrin (or brine) , 25% urethane was injected intraperitoneally to anesthetize ,all rats were recordⅡECG; drew blood from abdominal aorta to test blood lipid, hem- orheology and myocardial enzymes;got the myocardium to put up on electron microscope detect.Results:1 The dyslipidemia and myocardial ischemia rats model were established triumphantly.2 Tongmaihuazhuo decoction can reduce rats, blood serum TC and LDL-C obviously,and hoist blood serum HDL-C slightly.3 Tongmaihuazhuo decoction can reduce the whole blood and plasma viscosity of rats,.4 Tongmaihuazhuo decoction can reduce the level of CK,AST,LDH ,prove myocardial ischemia on ECG , prove myocardium ultrastructure of rats, and protect myocardial ischemia on a certain extent.Conclusion: Tongmaihuazhuo decoction can obtain the objective of lipid-lowering and lighten myocardial ischemia by the way of dissipating phlegm and removing blood stasis,that has the fuction of prevention and treatment to CHD.
