Different Emetogenic Substances Affect The Expression Of Sp And 5-ht In Rat Duodenal Tissue

OBJECTIVE To study the changing of SP and 5-HT levels in duodenum when rats were disposed by different medicine that can make people vomit and provide experimental basis for detecting the pathological mechanism of reverse ascending of Stomach—Qi.METHOD 1.Forty healthy Wistar rats were randomly divided into five groups:control 1 group, hellebore group,copper sulfate group,control 2 group, cisplatin group, 8 rats per group.The control 1 group were given saline via intragastric administration(i.g.), hellebore group were given concentrated solution of hellebore via (i.g.), copper sulfate group were given copper sulfate via (i.g.).The control 2 group were given saline via intraperitoneal injection(i.p.),cisplatin group were given cisplatin via (i.p.).6 hours later, duodenum was taken out of rats.The morphological changes were observed by HE staining.SP levels were measured with radioimmunoassay,5-HT were observed by high efficiency liquid chromatography,the expression of SP and 5-HT were detected by immunohistochemistry technique.2.The date were expressed as x±s.Independent-samples T test of SPSS10.0 was employed in the group comparion.RESULTS 1.The content of SP in the duodenum of copper sulfate group is more than those of the control 1 group;The content of 5-HT in the duodenum of copper sulfate group is more than those of the control 1 group. 2.The content of SP in the duodenum of cisplatin group is more than those of the control 2 group;The content of 5-HT in the duodenum of cisplatin group is more than those of the control 2 group;3.There is no significance of SP and 5-HT concentration between hellebore group and control 1 group.4.There are some inflammatory cell in copper sulfate group and cisplatin group.CONCLUSION1. Copper sulfate increase SP and 5-HT levels in the duodenum2.One of mechanisms of flaring up of stomach fire is associated with the increasing of SP and 5-HT levels in the duodenum.3.Cisplatin increase SP and 5-HT levels in the duodenum.4. One of mechanisms of YANG deficiency of the spleen and stomach is associated with the increasing of SP and 5-HT levels in the duodenum.
