Raspberry Study Chemical Composition And Quality Standards

Fructus rubi,the dry unfledged fruits of Rubus chingii Hu(Fupenzi) has been used as a Chinese herb in nourishing kidney.The previously studies showed that Fructus rubi mainly contained triterpenoids,volatitle oils,flavones,and otherwise.In present study,the compounds of Fupenzi were chemically studied systematically.From the 95%and 50%ethanol extracts of Fupenzi,18 compounds were isolated with modern chromatography methods,and among them,5 triterpenes, 3 alkaloids,4 flavones,2 sterols,and 2 other types of compounds were identified respectively based on spectroscopic and other chemical evidences.Among them,two ones were new found chemicals;another was new inartificial one;and the other four ones were isolated from the plant firstly.These compounds were identified as follows:Triterpenes:2α,19α,24-trihydroxyurs-12-ene-3-oxo-28-acid(1),tormentic acid (8),2α-hydroxyursolic acid(6a),maslinic acid(6b) and ursolic acid(7);Alkaloids:4-hydroxy-2-oxo-1,2,3,4-terahydroquinoline-4-carboxylic acid(2), Methyl 1-oxo-1,2-dihydroisoquinoline-4-carboxylate(3),1-oxo-1,2-dihydroisoquino line-4-carboxylic acid(4);Flavones:tiliroside(8),kaempferol(9),keamferol-3-O-β-D-glucopyranoside(10), quercetin(11);Sterols:β-sitosterol(12),daucosterol(15):Other types:triacontanol(13) and exadecanioc acid(14).Secondly,the HPLC method for determination of the content of tiliroside in the Fupenzi was established and 18 samples collected from different provinces were assayed.The result showed that method was simple,accurate and could be used for the quality control of the herb.Furthermore,the fingerprint of Fupenzi by HPLC was detected,and 10 common peaks areas in fingerprints(one of them was confirmed as the isolated compound mentioned above) were used for the calculation of similarity by the analytical software.The calculation result showed that there was no distinguished difference among the 10 samples from different provinces.And also,the TLC was studied for the discrimination of sample,and the content of water and ash were determined in the crude drug.And,the quality standard draft and related specifications were made on the basis of above studies.
