Rubi Parvifolii Radix is the dried root of Rubus parvifolius L. (Rosaceae), which has effects of resolving stagnation, clearing blood heat, killing pain, diuresis and detumescence, detoxication, antibacterial, etc. Radix Rubi Parvifolii as a commonly-used medical plant in folk, has promising therapeutic effect on gynecologic hemostasis, coronary atherosclerotic heart disease, hepatitis and so on. However, the chemical constituents and quality standard of Radix Rubi Parvifolii both could be further studied. In order to clarify the effective substance, establish quality standard of Radix Rubi Parvifolii, and provide reference for rational-and-safe use of crude drug and patented herbal medicine of Radix Rubi Parvifolii, systematical studies of the chemical constituents and quality standard of Radix Rubi Parvifolii were conducted in this project.31 compounds were isolated from the Rubi Parvifolii Radix with liquid-liquid extraction, silica gel, reversed phase chromatography, sephadex LH-20 column chromatography, macroporous resin column chromategra-phy and semipreparative HPLC. These compounds were identified on the basis of chemical and spectral methods. Two new compounds were parvif-olactone A (9) and Rubuside P (22).29 known compounds were p-hydroxybenzoic acid (1),4-hydroxy-3,5-dimethoxybenzoic acid (2),3-methoxy-4-hydroxy benzoic acid (3), β-sitosterol (4), oleanolic acid (5), ursolic acid (6),2-oxopomolic acid (7), pomolic acid (8), 18,19-seco,2a,3a-dihydroxyl-19-oxo-urs-11,13(18)-dien-28-oic acid (10), euscaphic acid (11), p-hydroxyphenethyl alcohol (12), maslinic acid (13), psiguanin A (14),2α-hydroxy ursolic acid (15), 1β-hydroxyeuscaphic acid (16), tormentic acid(17),2α,3α,19α,23-tetrahydroxyurs-12-en-28-oic acid (18),2α,3α,19α,23-tetrahydroxyolean-12-en-28-oic acid (19), L-epi-catechin (20),2α,3α,19α,23-tetrahydroxyurs-12-en-28-oic acid (21), glucosyl pinfaensate (23), Rubuside J (24),2α,3α,19α,24-tetrahydroxyolean-12-en-28-oic acid (25),2α,3α,19α,24-tetra-hydroxyurs-12-en-28-oic acid (26),2a,3a,19a,23-tetrahydroxyurs-12-en-24,28-dioic acid (27),2α,3β,19α-trihydroxyurs-12-en-23,28-dioic acid (28),2α,3β,19α-trihydroxy olean-12-en-23,28-dioic acid (29), suavissimoside Rr (30), daucosterol (31). Compounds 14, 25,26,29 were isolated from the family Rosaceae for the first time. Compounds 7,12 were isolated from the genus Rubus for the first time. Compounds 1-3,5,8,10,13,16-17,19,21, 23-24,27 were isolated from R. parvifolius for the first time.According to 2010 edition of Chinese Pharmacopoeia and obtained chemical constituents information, systematical study of quality standard of Radix Rubi Parvifoliix was accomplished. Origin, characters, macroscopical identification, TLC identification, inspection items, content determination and other indicators are included in the quality standard we established. With these multiple indicators, a more rational and safe medication of Radix Rubi Parvifolii crude drug and patented herbal medicine is expected.
