Rhizoma Atractylodis macrocephalae, the dried rhizoma of Atractylodes macrocephala Koidz., belongs to Compositae. with the efficacy of strengthing spleen and replenishing Qi, eliminating dampness, promoting urination, hidroschesis, calming fetus and anti-tumour action. As a traditional drug, Rhizoma Atractylodis macrocephalae has a long medicinal history and has been recorded by every China Pharmacopoeia. However, it has no quality standards and reference substance which was used for the qualitative identification and determination methods.In this paper, the anti-tumor action and the selection of candidates reference substances were studied.This paper can be divided into four parts:PartⅠ, the review of the study of chemical constituents, pharmacology and processing of Atractylodes macrocephala.PartⅡ, study of the chemincal constituents of Atractylodes macrocephala. This part can be divided into four sections:Section A: The extraction and separation of the volatile oil of Atractylodes macrocephala and identification of the chemical constituents. Three compounds were obtained from volatile oil of Atractylodes macrocephala Koidz.: atractylenolideⅠ, atractylenolideⅡand atractylenolideⅢ. These three compounds are all befitting candidates of the reference substaces.Section B: Study on the chemical constituents of aqueous extract of Atractylodes macrocephala. Scopoletin and scoparone were separated and identificated for the first time in Atractylodes macrocephala.Section C: Study on the chemical constituents of volatile oil of Atractylodes macrocephala by GC-MS qualitatively. The MS of each peak in TIC was obtained and 19 compounds were affirmed by comparison with references and the standard spectrum. The relative content of each compound was determined by area normalization.Section D: A rapid method combining gas chromatography with selecteive ion monitoring-mass spectrometry was established for the determination of atractylenolideⅠand atractylenolideⅡin volatile oil of Atractylodes macrocephala Koidz. with two candidates of the reference substaces as the standard substances. The determination method of volatile oil of Atractylodes macrocephala was established for the first time.And this determination method laid solid foundation to the supplement of Rhizoma Atractylodis macrocephalae in China Pharmacopoeia and the new antineoplastic drug development.PartⅢ, preliminary study on antineoplastic action of the constituents and volatile oil of Atractylodes macrocephala against S180 sarcoma and gastric sarcoma in tumor-bearing mice. The results of anti-sarcoma S180 showed that atractylenolideⅠ, atractylenolideⅢ, zbo-2 and zbo-1 all showed low tumor growth inhibition rate. While high dosage of zbo-1 showed relative high tumor growth inhibition rate. So it can be inferred that the antineoplastic active ingredients may exist in the low polarity fraction of volatile oil of Atractylodes macrocephala Koidz..PartⅣ, study on the stability of volatile oil of Atractylodes macrocephala.This article mainly has conducted the innovative research in the following aspects:A. The preliminary affirmation of effective fraction on anti-sarcoma S180 of volatile oil of Atractylodes macrocephala.B. The separation and identification of the markers in Atractylodes macrocephala. (absent from China Pharmacopeia). C. The determination method of volatile oil of Atractylodes macrocephala was established for the first time.D. The preliminary study on the stability of volatile oil of Atractylodes macrocephala.E. Two active constituents: scopoletin and scoparone were separated and identificated for the first time in Atractylodes macrocephala.
