The extraction and separation of the active components-Polyscaccharides, amino acids,flavones and essential oil separated from AtractylodesMacrocephala Koidz(AMK)at Pingjiang in hunan were investigated.With the optimal conditions of extraction of flavones were firstobtained as follow: adding 16 times of 95% ethanol as much as theweight of AMK at 80℃for 2h by 1 time extraction. There were at least 17kinds of amino acids in AMK, the contents of amino acids were firstdetermined.With decolorizing by active carbon in amino acid of AMK,the optimal decolorizing conditions were obtained as follow:0.02 times ofactive carbon as much as solution volume at pH1.0~1.5 at 70~80℃for35 minutes. The optimal conditions of extraction of water-solublePolysaccharides were obtained as follow: adding 10 times of water asmuch as the weight of AMK powder at 80℃for 2h and two times. Withthe orthogonal experiment design of L9(34),the optimal colored conditionsof Coarse water-soluble Atractylodies Macrocephalse KoidzPolysaccharides (CAMP) were obtained as follow: adding 4mL sulfuricacid and 1.2mL 5% Phenol at 25℃for 20 minutes. CAMP were purifiedby denionzing, decolorizing, removing protein, dialysis and deposition inorder to get refined Atractylodies Macrocephalse KoidzPolysaccharides(RAMP). Compared the atractylodes rhizome oil,Pericarpium Citri Reticulate rhizome oil and their mixed samples' oilswere obtained by the mixed material-water reflux condensation and themain effective constituents were identified by GC-MS, show that usingthe mixed material-water reflux condensation, the extraction process tothe mixed essential oil is simple and available.This research provides scientific reference for the further study anddevelopment of AMK as a natural plant resources in hunan.
