Objective:The researches of Atractylodes on the gastrointestinal tract focused on the effects of Atractylodes decoction.However,the compositions of Atractylodes decoction was complex and their contents were not clear.This topic studied the contents of atractylenolide in the alcohol extracts from Atractylodes,and the effects of processing on the intestinal muscle.Preliminary studies on dose effect relationship between the content and efficacy were also investigated.It was of great significance to the further studies of tractylodes.Methods:In this study,HPLC-DAD was developed for the determination of the three atractylenolide in the extracts of Atractylodes.The extraction and processing methods were optimized.The effects of processing on the chemical compositions of volatile oil from Atractylodes were studied by GC-MS.Excursive intestines flesh experimentation was carried out in order to compare the efficacy of three atractylenolide.We also studied the effects of processing on intestinal muscle tension.Results:?The optimum extraction conditions were determined with using 6 times of methanol and 8 hours each time.?The optimum processing conditions were determined with 10 times honey bran fried 10 min with 100?.After processing,the contents of atractylenolide ??? were 709.8 ?g/g(n=3,RSD=0.89%),193.1 ? g/g(n=3,RSD=1.47%)and 1174.1 ?g/g(n=3,RSD=1.34%),respectively.The extraction yields of volatile oil was 0.452%before processing,and 0.437%after processing.Four new components were found and five components disappeared in the volatile oil after processing.?It was found that two alcohol extract of Atractylodis could inhibit intestinal muscle tension.Processing could enhance the inhibition ability of Atractylodis extract.When the concentration of Atractylodis was 4.0mg/mL,intestinal muscle was almost completely inhibited.The inhibition ability of was stronger than atractylenolide ???.The inhibition ability of two alcohol extract of Atractylodis was almost same as atropine with 1/1000 concentration.Inhibition ability were alcohol extract of processing Atractylodis>alcohol extract of Atractylodis>atropine with 1/1000 concentration.Conclusion:In this study,quantitative analysis method was developed for the determination of the three atractylenolide.The extraction and processing methods were optimized.The effects of processing on the chemical compositions of Atractylodes were studied.In vitro intestinal muscle exercise test were carried out in order to compare the efficacy of alcohol extract of Atractylodis before and after processing.The project provided insights and technical supports for the innovation of discovering bioactive constituents in A tractylodes.
