Objective:Establishing the model of rabbits with vertigo of phlegm -retention syndrome type by feeding the high lipid diet and injecting tissue sclerosing remedy into the soft tissue on the left side of cervical vertebral transverse protuberance.To evaluate the success of the model and discuss mechanism of vertigo of phlegm-retention syndrome type,the experiment observed Levels of blood lipid,the vestibular blood supply and the duration of nystagmus.At the same time,the experiment observed the influence of Guben Huatan Dingxuan Decoction(GBHTDXD)to the model to explore the effect and mechanism of this therapy in treating Rabbits with Vertigo of phlegm-retention Syndrome Type.Methods:Forty normal rabbits were randomly divided into five groups: normal control group,untreated group,high-dose GBHTDXD-treated group, low-dose GBHTDXD-treated group and Sibeline-treated group.High fat diet were given to untreated group,high-dose GBHTDXD-treated group,low-dose GBHTDXD-treated group and Sibeline-treated group,and tissue sclerosing remedy were inject into the soft tissue on the left side of cervical vertebral transverse protuberance twice at the third and fourth week. Through this methods,the model of rabbits with vertigo of phlegm-retention syndrome type has been set up by lower the blood flow basilar arteriral arteries.From the ninth week,normal diet were given to every group.The low-dose GBHTDXD-treated group were given GBHTDXD at 6g/kg·d to affuse stomach,high-dose GBHTDXD-treated group were given GBHTDXD at 12g/kg·d to affuse stomach,Sibeline-treated group were given sibeline at 0.93g/Kg to affuse stomach,and the untreated group were given saline,while no treatment was given to untreated group.After two weeks' treatment,levels of blood lipid and were compared among the 5 groups. The microstructure of the vertebral artery were observed.The expressions of were NF-κB in Vertebral artery tested by immunohistochemical staining method.The blood supply of vertebro-basilar artery were tested by CDFI. And the duration of nystagmus were observed.Results:Comparing with normal group,the blood lipid level of TC,TG and LDL-C in untreated group have significant difference(p<0.01).The velocity in vertebro-basilar artery,the duration of nystagmus,and the expressions of NF-κB showed statistical differences between the normal control group and the untreated group(p<0.01).The microstructure of vertebral artery in untreated group were observed by means of HE,the vascular intima get thicker with a lot of foam cells and lipidosis.Comparing with model group,the level of TC,TG,LDL-C were much lower (P<0.01)or(P<0.05)in both of the low-,high-dose GBHTTMR-treated group.The pathological changes in vertebral artery has been delayed,the expressions of NF-κB(P<0.01)are less and increase the duration of nystagmus(P<0.05)were prolonged,and improvement was detected on blood viscosity in the high-dose GBHTTMR-treated group.The GBHTTMR of low-dose was non-effective in improving the duration of nystagmus and the vestibular blood supply(P>0.05).Sibeline can prolong duration of nystagmus(P<0.05),and increase the Vs and Vm of vertebro-basilar artery blood supply(P<0.01)or(P<0.05),but no significant changes in levels of blood lipid(P>0.05).Comparing with the low-dose GBHTTMR-treated group,TG elevates had the difference(P<0.05),LDL-C reduces had the remarkable difference (P<0.01)in the high-dose GBHTTMR-treated group.There is also a difference(P<0.05)of the expressions of NF-κB between them.Comparing with the Sibeline-treated group,the high-dose GBHTTMR-treated group had the remarkable differences in Vd of RVA,BA (P<0.05). Conclusions:The blood lipid level of TC,TG and LDL-C has an obvious increase in untreated group.The flow of vertebro-basilar artery is decelerated which shows a shortage of blood perfusion in BA,and the the duration of nystagmus was shortened which shows the sensibility of vestibular systems diminished and the function of left semicircular canal has been impaired.All of these indicates that the model of rabbits with vertigo of phlegm-retention syndrome type was set up successfully.GBHTDXD can decrease the blood lipid level of TC,TG and LDL-c,adjust the metabolism of blood lipid,and inhibit the expressions of NF-κB and the formation of AS.GBHTDXD may improve the blood flow of vertebro-basilar artery by reinforcing the compliance and diastolic-systolic action of vertebro-basilar artery.The effects of GBHTDXD may also depend upon activating the local microcirculation in the inner ear and improving the sensitivity of hair cells inside the cochlea, so as to recover the vestibular function and vestibulo-ocular reflex. Comparing with the Sibeline-treated group,the high-dose GBHTTMR-treated group improve the Vd of RVA,BA more obviously.
