Meridians And Kidneys, And Mechanism Of The Compound On The 5/6 Rat Nephrectomy Model Of Renal Protective Effect

ObjectiveTo study the pathogenisis and the foundation of the theory --"obstruction of kidney collaterals by blood stasis",and alse to clarify the mechanism of action that the Tongluo Baoshen decoction(TLBS) retards renal fibrosis,we use the 5/6 nephrectomy rats as model, and expect to find the theoretical base for anti-renal fibrosis by conbined treatment of traditional Chinese medicine and western medicine.Method1. 40 male Wistar rats, weighting 180-220g, adopted for a week. the rats were divided into 5 groups,which are normal group,model group, treated by Valsartan group,treated by Baoshenkang group and therapy group with TLBS decoction (n=8). All of the rats except the normal group were operated for 5/6 nephrectomy.Then rats of model group were fed with normal Saline,While other groups were fed with valsartan,Baoshenkang and TLBS decoction.We measured the weight of each rat every two weeks after operation,and we collected the Twelve-hour urine of every rat, recorded the UPD of each rat and tested the proteinuria at 4weeks and 6weeks after operation. We kill the rats at 6weeks.Blood urea nitrogen(BUN) ,serum creatinine,TG,CHO and LDL were measured before the rats were killed . Then we took the rats'kidneys into 10% neutral-buffered formalin to be fixed for pathological histological observation.The part samples were embedded in paraffin and steined with hematoxylin and eosin,PAS.2. Fn,Ln in the protein,collagen typeⅠ,Ⅲof rat's renal tissue was studied by means of immuno-histochemistry,and VEGFmRNA was study by hybridization in situ.3. SPSS13.0 was used.Data were analyzed using one-way ANOVA.All values were expressed as mean±SE,statistical significance was assigned at a p value of <0.05 or <0.01.Result1. The TLBS decoction can decrease the loss of rat's urine protein and the production of BUN and SCr,protect the renal function,improve the pathological lesion of renal tissue,protect the remaining nephron, retard renal fibrosis2. The TLBS decoction can level down the content of Fn,Ln, collagen typeⅠ,Ⅲ,so that it can inhibit the accumulation of ECM,retard renal fibrosis.3. The TLBS decoction can up-regulat the expression of VEGFmRNA, which means that one of mechanisms could be it.Conclusion 1. we consummate the theory of obstruction of kidney collaterals by blood stasis,bring forward the TCM chief pathogenisis,which has three stages,there're exopathogen invading the collaterals,toxin invading the kidney collaterals, accumulation of toxin and blood stasis.we also bring forward using the model of renal fibrosis as a method to investigate the rationale and treatment prescription,and the accumulation of ECM embodiment the"pathogen usually intrudes into collaterals in protracted disease, accumulation of toxin and blood stasis".2. The TLBS decoction can retard renal fibrosis in many different ways,and it can treate renal injury in early stage of CKD.
