Purpose:Diabetic cardiomyopathy (DC) is a specific cardiomyopathy, pathological changes mainly as cardiomyocyte hypertrophy, degeneration, focal necrosis, lost a lot of muscle fiber filaments, myocardial extracellular matrix accumulation have insoluble collagen fibers technology. Myocardial collagen I, III (Col I, Col III) is the main component of myocardial constituted, has close ties with myocardial fibrosis. This study was to investigate the effect of acupuncture on experimental diabetic cardiomyopathy and investigate the influence of myocardialcollagen I, III (Col I, Col III) expression levels. Which can further reveal the mechanism of acupuncture treatment of DCM, to provide experimental evidence and theoretical basis for acupuncture treatment of DCM.Method:1. Modeling Methods:70healthy male SD rats were fed for one week, checkblood sugar normal, qualitative urine test was negative, the animal is in good health who wererandomly divided into two groups, namely the control group10DCM model production team60Normal control group fed with normal diet always, model-making group55mg/kg dose byintraperitoneal injection (ip)1%STZ, After injection STZ72h, cut tip of the tail, fastingplasma glucose and qualitative urine, fasting plasma glucose≥16.7mmol/L, urine≥+++,included in the model replication group. Successful modeling51(85%success rate modeling,died two) were randomly divided into acupuncture group17,17drug treatment group, modelcontrol group17.2. Treatment: drug treatment group, according to the human rat body weight conversionformula fed daily morning regularly crawl enalapril15.mg/kg; normal control group andmodel control group fed an equal volume of drinking water daily morning; acupunctureacupuncture treatment group rats Feishu, spleen, Shenshu, Zusanli, Sanyinjiao five acupoints,acupuncture every other day tied once every30minutes;12weeks after cardiac death whiletaking in the rats prepared for testing.3. Blood, urine and weight measurement: the use of glucose meters in rats fasted for12hours, the tip of the tail fasting blood glucose was measured in rats; simultaneously conductqualitative and weight of the rat urine of rats, a week once.;4. Cardiac index determination: After the rats were killed, weighing his heart, left ventricular mass, calculating mind body ratio index (H/B, heart weight/body weight) andleft ventricular mass index (LVMI, left ventricular weight/body weight);5. DCM rat myocardial collagen I, III immunohistochemical analysis: rat heart tissueslices, immunohistochemical staining. Compare rats myocardial collagen I, III proteinstaining;6. Statistical analysis: SPSS13.0statistical software for statistical analysis using t testbefore and after treatment, count data (X±s) said.Result:1. After treatment, the DM rats in the acupuncture treatment group comparedwith the model group had a to a significant difference (P <0.01) between blood glucose, bodyweight and the clinical features;2. The H/B and LVMI between two groups had a significant difference (P <0.05,P<0.01);3.The myocardial collagen protein I, III expression levels Rats in the acupuncturetreatment group were significantly reduced compared with the model group (P <0.01);4. Both intervention therapy on myocardial collagen I, III in two groups of DM ratsshowed no significant difference (P>0.05) level of regulation.Conclusion:1, Acupuncture DM rats’ Feishu, Pishu, Shenshu, Zusanli, Sanyinjiao canregulate their blood sugar levels, urine sugar levels, can significantly improve the generalstate of DM rats;2, Acupuncture can significantly improve diabetic rat heart H/B and LVMI, can protectrats’myocardial;3, Immunohistochemistry results suggest that myocardial collagen I, III involved in thepathogenesis of DCM; acupuncture DM can inhibit myocardial collagen I, III expression, andits result is similar with enalapril;4, Acupuncture can effectively improve the prevention and treatment of DCM, can treatthe diabetic cardiomyopathy. The mechanism is inhibit the level of myocardial collagen I, III’s expression.
