Abstract/Summary:
Objective: To investigate G. pentaphllum's influence on the content of hydroyproline (HYP) and Malondialdehyde (MDA) in the skin of hairless photo ageingmice as well as the activity of super oxide Dismutase(SOD) in their skin , and to investgate G. pentaphllum' s effect on the structure of hireless photo ageingmiceMethods: Healthy KM hairless mice were divided into seven groups randomly and gavages respectively,that is negative Control Group ( A ) , Model Group ( B ) , Model+Low-G. Pentaphyllum group ( C ) , Model+Medium-G. Pentaphyllum Group ( D ) , Model+High-G. Pentaphyllum Group (E). The mice in B, C, D, and E group were irradiated to simulate skin photoageing by long-term ultraviolet irradiation, at the same time , those in C, D, and E were given G. Pentaphyllum. Staining skin with HE, Under 40 times light microscope, the structure of dermal were observed. While the changes of the contents of dermal hydroxyproline and Malondialdehyde, as well as the activity of Superoxide Dismutase were measured by biochemical test method in each group of the experiment.Results: In model group, the activity of SOD and the content of HYP of dermal was decreased, while the content of the MDA was increased seriously (P<0.05) The skin biopsies appear photoageing look. The contents of SOD and MDA and HYP in dermal in group C, D, E had no difference with that in group A. the activity of SOD in dermal in group C, D, E was increased (P<0.05 ; P<0.01) . while the content of MDA was reduced in group C, D, E . The contents of activity in the activity of HYP in dermal in group D, E was increased (P<0.01; P <0.05) .Clusions: G. Pentaphyllum has the protective effect on the skin of the photoageing hairless mice with ultraviolet irradiation. The mechanism may be that on one hand, G. Pentaphyllum could eliminate free radicals itself; on the other hand, by increasing the activity of SOD and HYP, it could inhibit the oxidative damages of ultraviolet and skin photoageing.
