Objective: To investigate Poria's influence on the content of hydroxvproline(HPY) in the skin of aged rats as well as the activity of Superoxide Dismutase (SOD) in both rat's skin and red blood cells, and to infer Poria's function and mechanism of rotarding the senility of skin. Principle: This experiment is based on the theory of spleen deficiency-skin senility and Poria's effect in Traditional Chinese viedicine, Free Radical Theory and Cross-linking Theory in modern medicine as well. Materials and Methods: Female Wister rats were divided into six groups randomly and gavaged respectively, that is Young Control Group (NS 20ml/kg), Old Control Group (AS 20ml/kg), Positive Control Group (Vitamin E2g/kg), Low-Poria-Dosage Group (Poria 2g/kg), Medium-Poria-Dosage Group (Poria 4g/kg), High-Poria-Dosage Group (Poria 8g/kg). Peria, a kind of spleen-reinforcing herbal, is used in this study. After 33 days gavages, all the rats were killed to detect the changes of activity of SOD in both rat'ss skin and red blood cells and the content of HPY in rat' s skin in all groups. Results: The content of HPY in the skin of Young Control Group, Positive Control Group and High-Poria-Dosage Group are higher than that of Old Control Group (p<0.05). The activity of SOD (both in red blood cell and in skin) in Poria Groups have no great changes compared with that of Old Control Group (p>0. 05). Conclusions: Poria probably have the function of increasing the content of HPY in the aged rats' skin while it has no influence on the activity of SOD in red blood cell or derma of aged rats.
