Objective:To study the mechanism of anti-aging effect of dimethylaminoethanol (DMAE) and Compound Amino Acid(AA) injection by Mesotheray in D-galactose-induced chemical skin aging rat.Methods:Eighty rats were randomly divided into a treatment group of aging (60), the aging control group (10) and normal control group (10). The aging treatment group, aging control group rats were injected subcutaneously with D-galactose125mg· kg-1. d-1during42days. From the18th day, the aging treatment group were injected intradermally in the rats’both sides hip skin with0.2%DMAE+AA,0.1%DMAE+AA,0.2%DMAE,0.1%DMAE, AA, saline,respectivly, once a week. After four weeks, HE, water content, Super oxide dismuitase (SOD) activity, hydroxyproline (HYP) and malondialdehyde (MDA) content were measured.Results:Compared with the aging control group, the epidermal thickness, the dermal thickness and the density of Collagen fibre of rats’ skin in aging0.2%DMAE+AA and aging0.1%DMAE+AA treatment groups increased significantly (P<0.05). SOD activity and HYP of aging0.2%DMAE+AA and aging0.1%DMAE+AA treatment groups were significantly higher than other groups(P<0.05),but their MDA was obviously lower than that of other groups. There were no difference among normal control group,aging0.2%DMAE+AA group and aging0.1%DMAE+AA group. There were no differences in water content among groups (P>0.05).Conclusion:Intradermal injection with0.1%DMAE+AA and0.2%DMAE+AA in aging rats’ skin may increase the epidermal thickness, the dermal thickness and the density of Collagen fiber of rats’ skin, improve the activity of SOD, increase HYP content and decrease MDA content, indicating that it has a ability to scavenge skin free radicals, enhance antioxidant capacity and to postpone skin collagen metabolism, which can prevent skin aging.
