Investigation background Synovitis is the distinctive pathology of rheumatoid arthritis (RA). Vegetation and pathology of synovium are similar to tumor in many ways. Matrine(Mat) is the main alkaloid of foxtail-like sophora herb and seed and one of newfashioned antitumor drugs. We presume Mat could be employed in the treatment of RA.Objective To investigate the effects of Mat on cell proliferation, PCNA and cell cycling of the Henrietta Lacks strain of cancer cells(Hela cells) from growing in vitrocolture environment. Furthermore, to compare with methotrexate (MTX) and provide scientific basis for Mat employed in the treatment of RA.Method The level of cell proliferation was measured by the MTT method, cell cycle was measured by flow cytometry with PI Staining the flow cytometry (FCM) method, and the level of proliferating cell nuclear antigen (PCNA) was measured with immunohistochemistry(IHC).Result Level of proliferation of Mat-treated (0.5 to 1.5g/L) and MTX-treated (2.5 to 25mg/L) Hela cells was significantly decreased compared with corresponding not-treated group(P<0.05). Both drugs inhibited proliferation of Hela cells in a dose-dependent manner. FACS assay showed the rate of MTX-treated and Mat-treated Hela cells in G1 phase as significantly increased than not-treaded group(P<0.05), as well as the rate of Hela cells in S phase and G2 phase(P<0.05). Level of PCNA of Mat-treated and MTX-treated groups dropped significantly compared with control group(P<0.05). But these changes were roughly similar between MTX-treated and Mat-treated Hela cells.Conclusion The findings suggest that Mat inhibites the proliferation and proliferation-dependent processes of Hela cells, which was similar to MTX. Mat has the potential value in the treatment of RA.
