Blood Deficiency (BD) is common in the clinic of Traditional Chinese Medicine, however there are few reports about the BD model which closely resembles the clinic BD.The research is based on the two models of mouse established by using (60)Coγ irradiation or cyclophosphamide (CTX) and sponsored by the National Natural Science Foundation Project of China which are 《The Study of Molecular basis of BD of Chinese Medicine》and 《The Basic Study of Composition Theory of Siwu Tang》.In order to investigate the involvement of changes of hematopoietic cells during the process of BD and evaluate the rule of disease development of Siwu Tang (SWT), we built the blood deficiency models by limiting diet, swimming and chronical bleeding. Then, the numbers of circulating blood cells, the percentage of (CD34)+ cells in bone marrow, the apoptosis, the cell cycle and the ability of colony-forming unit of bone marrow cells have been observed. SWT (Decoction of four ingrendients), composed of Radix rehmanniae, Radix angelicae sinensis, Radix paeoniae alba and Rhizoma chuanxiong, is a famous traditional Chinese medical formula for enriching and regulating the blood. The molecular mechanism of SWT on the BD mice made by compound bleeding (CB) have been studied. By feeding diet according to 75g.kg-1 weight every day, swimming 20 minutes and removing 0.4ml blood every mouse every two days, we built the blood deficiency models. The main results are as follows: At the ninth day of making model, the indications of BD including decrease of peripheral blood cells, indolence in action and wizened fur appeared significantly in mice. Compared with normal group, the total numbers of RBC and HGB in blood decreased significantly from the day of making model, and the rate of (CD34)+ cells in bone marrow cells decreased at the 7th and 12th day in the experimental group. At the 4th, 7th and 21st day, the numbers of cell of G0/1 cycle reduced and those of S cycle increased significantly respectively. In addition, the index of thymus decreased significantly. The whole blood decreased and then the numbers and capacity of hematopoietic cells proliferated quickly after incessant bleeding which destroyed the homeostasis of hematopoietic stem and progenitor cells in bone marrow. To study the effects of SWT on BD mice induced by CB, SWT was given to mice by oral administration before or after the establishment of the model , respectively. Oral administration of SWT when begin building model can promote proliferation of bone marrow cells, decreasing apoptosis, increasing immunity of BD mice. Promotion on recovering of stem and progenitor cells in bone marrow and the total numbers of peripheric blood, decrease the block of bone marrow cells in G2/M cycle and mobilizing more cells into S cycle and decrease of apoptosis were obviously seen in the preventive group of mice. The function of promoting the capacity of proliferation and decreasing apoptosis of hematopoietic stem and progenitor cells of SWT was thought to contribute to its influences on the mice of BD induced by CB. The compared experiment of BD built by different ways which are CB, (60)Coγ irradiation and CTX shows the result as follows: the total numbers of RBC and HGB reduced in CB group, those of WBC decreased in (60)Coγ irradiation group, and those of RBC and WBC lowered significantly in CTX group. The numbers of CFU-E,BFU-E,CFU-GM reduced evidently in three groups at the 15th day and after 7 days respectively. The numbers of (CD34)+ cells were reduced remarkably in (60)Coγ irradiation and CTX group while increased markedly in CB group. In addition, the index of thymus of all three groups lowered than normal group repectively. In sum, aimed at reducing the numbers of cells in peripheric blood directly, compound bleeding is more near the BD in clinic, and the method to build model possesses the virtue of simple way and clear index.
