Objective: Through the effect of spleen deficiency on the aquaporinS of the epithelial cell in the colon, try to find out the physiological role of aquaporins in the colonic fluid metabolism, and further provide the experimental evidence of the occurrencing mechanism of spleen deficiency diarrhea.Method: Using the binding methods of diarrhea of bitter and cold, abnormal of starvaion and full and excessive labor to create the rat model of spleen deficiency. Observe the character of the rats defecation, body weight and eating weight to determine the model are successful or not; Using the method of HE staining observe the state of colonic mucosa ; Using the method of reverse transcript of polymerase chain reaction (RT-PCR) to detect the mRNA expression quantity of aquaporinS in the colon; Using the method of immunohistochemistry to detect the protein expression quantity of aquaporinS in the colon. Compare the change states of spleen deficiency group and the normal group.Result: Compared to the normal group, the rats of spleen deficiency had the evident symptom of diarrhea , body weight grew slowly(P<0.05) and eat less(P<0. 05). HE staining of colonic sections under the light microscope displayed: colonic mucosa were not integrated, some epithelial cells of top villi abscised, cells arrayed scarcely; the mRNA and protein quantity of aquaporinS in colonic epithelium of spleen deficiency rats were all less than that of the normal group(P<0.05).Conclusion: 1, Spleen deficiency can cause the colonic mucous epithelial cell to abscise and effect the integrity of colonic mucous epithelium.2, Spleen deficiency can cause the expression of mRNA of aquaporin8 in the colon to decrease evidently.3, Spleen deficiency can cause the expression of protein of aquaporin8 in the colon to decrease evidently.So,we can conclude from above that Aquaporin8 may have important physiological role in the colonic reabsorption of water and reduction of aquaporin8 may be one of the pathological mechanism of spleen deficiency diarrhea.
