ObjectiveYunPiZhiXie herb by the late famous Traditional Chinese Medicine(TCM)pediatrics expert professor wang nationwide for pixu(spleen deficient)established by the etiology and pathogenesis of diarrhea of prescriptions,the whole party by nutmeg,cloves,poria cocos,red alloysite,FuLong liver,cold water stone,lotus seed,malt,divine comedy and twelve taste of traditional Chinese medicine(TCM),is clinically for protracted diarrhea,diarrhea type pixu(spleen deficient)with good clinical effect.Found in a previous study,the spleen antidiarrheal can improve pixu(spleen deficient)type of diarrhea of children with intestinal flora imbalance,reduce the amount of enterococcus,e.coli,to increase the number of bifidobacteria,bacteria and lactic acid bacteria,thus promote the digestion and absorption of protein,at the same time can slow gastric bowel movement,inhibit diarrhea,has an obvious protective function of small intestine microvilli.YunPiZhiXie herb has active ingredients such as volatile oil,silicic acid,alumina,etc.,which can promote gastric juice secretion,protect gastric mucosa,prevent diarrhea,antiviral,antimicrobial and regulate immunity and other extensive pharmacological effects to treat diarrhea by absorbing toxic by-products of digestive tract.This experiment intends to establish the model of persistent diarrhea of spleen deficiency in children,and explore the establishment standard of the model of persistent diarrhea of spleen deficiency in children,so as to provide a new idea for the establishment of the model of persistent diarrhea of spleen deficiency in children.At the same time,the intervention effect of Yunpi Zhixie Decoction on the model mice with persistent diarrhea caused by spleen deficiency was also clarified.The mechanism of Yunpi Zhixie Decoction in the treatment of persistent diarrhea caused by spleen deficiency was also clarified by observing the changes in the content of short-chain fatty acids in their feces.MethodsThis experimental study with"high concentrations of senna water decoction lavage" children of diarrhea of pixu(spleen deficient)model is set up,were randomly divided into normal group,model group,the spleen antidiarrheal party group and montmorillonite powder group,observation made in rats model and general situation changes after the treatment,weight gain,diarrhea,diarrhea index duration,using high performance liquid chromatography(HPLC)method to detect concentration of short chain fatty acids were feces.Results1.Weight change of young mice before and after modeling and treatment:Before modeling,there was no significant difference in body weight between normal group and model group(P>0.05).After modeling,the body weight and weight gain of the model group were significantly lower than those of the normal group(P<0.01).Shipped at the end of the treatment,compared with normal group,spleen antidiarrheal soup group of lower body weight(P<0.05),model group,the montmorillonite group was significantly lower(P<0.01);Group compared with model group,the montmorillonite powder weight is higher(P<0.01),YunPiZhiXie herb group was significantly higher(P<0.01);Compared with the montmorillonite group,YunPiZhiXie herb group had a significantly higher weight of diarrhoea(P<0.01).Compared with normal group,model group,the montmorillonite group weight growth was significantly lower(P<0.01),YunPiZhiXie herb group weight low growth of diarrhoea(P<0.05);Compared with model group,transporting spleen decoction group had a significantly higher body weight increase of diarrhoea(P<0.01);Compared with the montmorillonite group,YunPiZhiXie herb group had a significantly higher body weight increase of diarrhoea(P<0.01).2.Comparison between the duration of diarrhea and average diarrhea index of young mice in each group after modeling:During the period of treatment,the normal group diarrhea significantly shorter duration compared with the other three groups(P<0.01);Compared with model group,the montmorillonite powder group had a significantly shorter diarrhea duration(P<0.01),transporting spleen decoction group had a significantly shorter diarrhea duration of diarrhoea(P<0.01);The duration of diarrhea in the montmorillonite powder group was slightly shorter than that in YunPiZhiXie herb,but there was no statistical significance after testing.At the end of modeling,the diarrhea index of the normal group was much lower than that of the other three groups.With the progress of treatment,the diarrhea symptoms of the montmorillonite group recovered the fastest,followed by the YunPiZhiXie herb group,and the worst was the model group.The antidiarrheal effect of the western medicine montmorillonite powder was slightly better than that of YunPiZhiXie herb,but the test showed no statistical significance.3.After the end of treatment,the concentration of short-chain fatty acids in feces of young rats in each group:At the end of the treatment,compared with normal group,model group,the montmorillonite group acetic acid were significantly higher(P<0.01),model group of propionic acid were significantly higher(P<0.01),model group butyric acid were significantly lower(P<0.01),YunPiZhiXie herb group of butyric acid level is higher(P<0.05),model group 2-methyl butyric acid were significantly higher(P<0.01).Compared with model group,the acetic acid level of montmorillonite group and YunPiZhiXie herb group was significantly lower(P<0.01),the propionic acid level of montmorillonite group and YunPiZhiXie herb group was lower(P<0.05),the butyrate level of montmorillonite group and YunPiZhiXie herb group was significantly higher(P<0.01),and the 2-methylbutyrate level of montmorillonite group was lower(P<0.05).Compared with the montmorillonite group,the acetic acid level in YunPiZhiXie herb group was significantly lower(P<0.01),and the butyric acid level in YunPiZhiXie herb group was higher(P<0.05).Conclusion1.The model was established with the method of"high-concentration senna leaf water decoction by gavage".During the modeling process,the young mice gradually showed signs of spleen deficiency such as listlessness,reduced diet,activity,weight loss,etc.,and after the modeling,the young mice in the model group could still maintain symptoms of diarrhea,which better reflected the syndrome manifestations of prolonged diarrhea of spleen deficiency.Compared with other modeling methods of spleen deficiency diarrhea,this method can perfectly replicate the pediatric model of persistent diarrhea with spleen deficiency.2.The formula can effectively improve the symptoms of young mice with persistent diarrhea caused by spleen deficiency,and effectively improve their body weight and weight growth rate.At the same time,it can significantly shorten the duration of diarrhea in young mice model of persistent diarrhea caused by spleen deficiency,and the antidiarrheal effect of western medicine montmorillonite powder is slightly better than that of Yunpi antidiarrheal prescription.3.The formula can significantly reduce the concentration of acetic acid in the intestines of young mice,and at the same time increase the concentration of butyric acid in the intestines,improve the digestive and absorption function of the digestive tract,so as to improve the diarrhea symptoms of young mice with persistent diarrhea caused by spleen deficiency,and achieve the purpose of treating persistent diarrhea caused by spleen deficiency.
