Objective:To observe the therapeutic effect the main pharmacological action of Zhi Sou Gao(ZSG) by applying on the points (Dingchuan\Feishu\Tiantu\Tan zhong\Yongquan) on children patients in cough and cough modles; To study the antiviral and antibiosis action of ZSG on virus and bacteria by using cell culture technique. And to explore the mechanism of ZSG.Methods:On the clinical study:Nine hundred and eighty cases of cough were randomly divided into three groups,Group A (n=580) was treated with ZSG ;Group B (n=200) was treated with Amoxicillin capsules;Group C was treated with Ji Zhi Syrup .The changes of clinical symptoms signs and therapeutic effects were observed.On the experimental study:Firstly,the cough rat guinea pig rabbit models were established to observe the therapeutic effects (such as stopping cough, relieving asthma, dissolving phlegm) and the toxic and side reations with different doses of ZSGSecondly,the maximal non-toxic concentration of ZSG in Hep-2 cell culture was measured;the antiviral and antibiosis action of ZSG on the Respiratory syncytial virus(RSV) the Adenovirus(ADV) and five kinds of common bacteria were calculated;In addition,the degrees of inhibition of the drugs on the cytopathic effect of viruses in cell culture were observed by changing the concentrations of ZSGResult:On the clinical study:After treatment,in Group A:the cured rate was 50.69%;significantly effective rate was 69.48%;the total effective rate was 83.10%.The above indexes have no difference among three groups(P>0.05).The time of diet recovery CD3 CD4 CD8. IgA in Group A were superior to those in Group B and C(P<0.01).The differences of BUN ALT were insignificant in Group A before and after treatment(P>0.05).On the experimental study:The maximal non-toxic concentration of ZSG was 500 g/ml;ZSG inhibited the replication of RSV and ADV in cultured cells ,the degrees of cytopathic effect were different when changing the concentration of ZSG;To Staphylococcus, the minimum antibiotic concentration of ZSG was 0.5g/ml;To Steptoccaceae pneumoniae, it was 0.125g/ml;To a -Hemolytic strepococcus it was lg/ml;To Escherichia Coli it was 0.5g/ml;To Pseudomaonas Aeruginosa it was 0.5g/ml.The effects of ZSG to prolong the cough latent period and to stop cough were significantly different with the control group treated with nothing(p<0.05),the same as the effects to dissolve phlegm and relieve asthma.ZSG had no acute toxicity stimulation and hypersusceptibility reactions.Conclusion:The Traditional Chinese Medicine (TCM) prescription of ZSG applied on the points of the children patients in cough could alleviate the clinical symptoms,shorten the course,and the therapeutic effect was significant.And it also could obviously improve the function of immune.On the other hand,ZSG exerted a good effect in antiviral and antibiosis action. It was proved to be safe,easily operated ,economical and practical.
