Purpose:Through observation of acupuncture on experimental diabetic rat myocardium expression of MCP-1 ,acupuncture in the treatment of the disease to further explore the role and effectiveness.Method: 1 Modeling method: 40 healthy male Wistar rats by intraperitoneal injection of STZ, diabetic rats induced by experimental, 72h, based on the detection of fasting blood glucose level, were randomly divided into normal control group 8, acupuncture group, 10 rats with enalapril treatment group 11, model control group 11, DM 80% success rate modeling.2 Treatment: group therapy with enalapril daily morning under the weight of rats fed with enalapril 1.5mg/kg;Normal control group and DM model group fed with the same volume daily from drinking water; acupuncture acupuncture rats Feishu, spleen, Shenshu, internal relations, Sanyinjiao, epigastric Yu and other points under; time of 12 weeks.3 Detection of target: 14 week, 1) fasting blood glucose level testing; 2) body weight in rats before and after comparison; 3) HE Stained myocardial tissue; 4) immunohistochemical staining of myocardial tissue MCP-1 protein expression.4 Statistical analysis: The SPSS14.0 statistical software for statistical analysis, before and after treatment by t test with count data ( x±S) said.Result:After 14 weeks treatment, 1) acupuncture group FGB significantly lower than before treatment (P<0.01), with statistical significance;2) acupuncture group can improve the general state of DM rats;3) acupuncture group by light microscopy mainly as follows: myocardial fiber arrangement is slightly disordered, no gap widened, muscle fiber breakage, significantly improved compared with the model group;DCM model group, mainly for the light microscopy of myocardial fibrosis in interstitial cells, fibroblasts, enalapril treatment group and model group, the performance of similar, but better than the model group;4)immunohistochemistry DCM myocardial tissue MCP-1 results:Acupuncture experimental group and enalapril drug treatment in the control group MCP-1 expression in the myocardium from the area ratio of staining intensity to the DCM model group decreased significantly, semi-quantitative analysis showed that acupuncture group close to MCP-1 protein expression in the normal control group, acupuncture group and the enalapril experimental positive control group myocardial infiltration of mononuclear macrophages decreased, respectively, compared with the DCM models were significant statistical significance (P<0.01), but the two groups, the acupuncture group was better than enalapril positive control group.Conclusion: 1acupuncture rats Feishu, Pishu, Shenshu, internal relations, Sanyinjiao, epigastric Yu and other points can be adjusted under the fasting blood glucose levels, can improve the general state of DCM in rats;2) Acupuncture can improve the experimental DCM myocardial pathology of DCM has a protective role in heart; 3) molecular biology experimental results show that acupuncture is more inhibited DCM myocardial expression of MCP-1 protein role. Myocardial MCP-1 protein involved in the pathogenesis of DCM. Suggest the role of acupuncture control DCM myocardial tissue may be through inhibition of MCP-1 channel, the reduction of MCP-1 over-expression, inhibition of inflammatory DCM pathogenesis.
