Purpose: Diabetic cardiomyopathy is a specificity of heart disease caused by diabetesmellitus. Pathological changes in diabetic cardiomyopathy mainly in threeaspects.First,cardiomyocyte hypertrophy, degeneration and focal necrosis.Second,lost a lot ofmuscle fiber.The third, myocardial cell outer insoluble collagen accumulation and fibrosis. Inthis paper,we obtains from the key diabetic cardiomyopathy pathological changes ofmyocardial fibrosis, study of acupuncture effect on protein kinase expression in the themyocardium of experimental diabetic cardiomyopathy rats to further provide new discussesthe function and mechanism of acupuncture treatment of diabetic cardiomyopathy and theclinical diagnosis and determine the efficacy of diabetic cardiomyopathy to provide newtheoryetical and experimental basis.Method:1,Modeling method: choose70healthy male SD rats divided into two groups,ten rats in the normal control group, the others in the DM model group. Model groupaccording to the dose of intraperitoneal injection of55mg/kg (i.p.)1%STZ. After72hours,we determined fasting blood glucose and glycosuria test. The rats which fasting blood glucose≥16.7mmol/L, glycosuria test≥+++included in the model group.51rats were eventuallysuccessful model, these rats were randomly divided into3groups. There are17rats in theacupuncture treatment group,17rats in drug treatment group and17rats in model controlgroup (DCM model success rate of about85%, death,2);2,Treatment: drug treatment group, according to the conversion formula of human-mouseweight, daily morning lavage enalaprilat15mg/kg; Normal control group and model controlgroup fed with an equal volume of drinking water; Acupuncture treatment rats acupuncture5acupoints, there are Feishu,Pishu,Shenshu,Tsusanli and Sanyinjiao, on alternate days needle1times, each time30minutes.12weeks after each sampling at the same time detection;3,The measurement of blood glucose, glycosuria test and body weight: We measuredblood glucose after fasting12h, and do the glycosuria test and weight detection, are1perweek;4, Cardiac index determination:after the rats were sacrificed,weighing the whole heartand the ventriculus sinister, calculate the heart body than index(H/B, the entire heart heavy weight/body weight) and left ventricle mass index(LVMI);5, PKC in myocardium of diabetic rats with immunohistochemicalanalysis: slice heart tissue of rats, by immunohistochemical staining. Comparison of ratmyocardial PKC protein staining;6, Statistical analysis: The SPSS17.0statistical software for statistical analysis, usingsingle factor analysis of variance and nonparametric tests,with count data (xS) said.Result: After treatment1, compared with the model group,the diabetic rats inacupuncture treatment group blood glucose decreased and body weight increased significantly,with a significant difference (P <0.01);2, compared with the model group,the diabetic rats in acupuncture treatment group heartH/B and LVM reduced significantly, with difference (P <0.05,P <0.01);3, compared with the model group,the diabetic rats in acupuncture treatment group PKCexpression levels in myocardial of rats were reduced significantly, with a significantdifference (P <0.01);4, acupuncture treatment group and drug treatment group showed that both treatmentshad no significant difference (P>0.05) on the expression levels of PKC in myocardial of rats.Conclusion:1, acupuncture Feishu,Pishu,Shenshu,Tsusanli and Sanyinjiao not only canregulate the blood sugar and urine sugar levels,but also significantly improve the general stateof diabetic rats;2, acupuncture can significantly improve the diabetic rats heart of H/B andLVMI,and have some regulation and protection on myocardium of diabetic rats;3,immunohistochemistry showed, PKC involved in the pathogenesis of DCM; acupunctureinhibit PKC expression in diabetic rats myocardium, and its role similar effects on enalapril;4,acupuncture treatment of diabetic cardiomyopathy, can inhibit myocardium PKC expressionlevels play a therapeutic role in DCM, can effectively improve the prevention and treatmentof DCM.
