Background and objectiveOvarian Carcinoma is a one of the most common malignant gynecological tumors.Owing to its rapid growth and early abdominal dissemination,the long-term survival of patients is rather low,usually 30%.The pathogenesis of cancer is not identified now,but it is correlated with multiple gene mutation.Recently cell proliferation has been hot spot in the research of tumor.AnnexinⅡplays a very important role in DNA synthesis and cell proliferation.Expression of AnnexinⅡis significantly elevated in several human cancers.CyclinD1 is the oncogene which controls cell cycle.If expression of CyclinD1 is excess,it make that cell proliferation is out of control,which may induce cancerization.Ki-67 protein antigen is correlated with cell cleavage and proliferation,and is the sensitive index that reflects cell proliferation.But there was no report about the relations between AnnexinⅡand CyclinD1or Ki67 in ovarian carcinoma.This study will detect the expression of AnnexinⅡ,CyclinD1 and Ki67 protein in specimens of ovarian adenocarcinoma and ovarian adenoma by the technique of immunohistochemistry.And we will explore the relationships among them on the level of protein.We will provide theory and experiment evidence for treatment of ovarian carcinoma by inhibition of cell proliferation.Materials and methods48 cases of ovarian adenocarcinoma were collected from 2005~2006 year's file paraffin block in the First Affiliated Hospital,Zhengzhou University.All the cases had no history of the chemotherapy,radiotherapy and immunotherapy.All the specimens are graded according to WHO pathology standard.13 cases are well-differentiated,14 cases are moderately-differentiated,and 21 cases are poorly differentiated.All the specimens are staged according to FIGO of 2000 year.5 cases areⅠstage,10 cases areⅡstage,31 cases areⅢstage,2 cases areⅣstage.33 cases had the lymph node metastasis,15 cases non-lymph node metastasis.Other 10 cases are ovarian adenoma.The expressions of AnnexinⅡ,CyclinD1 and Ki-67 protein were detected in 48 cases of ovarian adenocarcinoma and 10 ovarian adenoma by immunohistochemistry. All the data were analyzed by SPSS10.0 statistical package.The comparison of positive rate uses the Chi-square;the relation of two variables is analyzed by Spearman's correlation analysis.The level of significant difference was a=0.05.Results1.The positive rates of expression of AnnexinⅡprotein in ovarian adenoma were 50.0%(5/10),and in ovarian adenocarcinoma were 81.3%(39/48).There was a significant difference between ovarian adenocarcinoma and ovarian adenoma (P<0.05).2.The positive rates of expression of CyclinD1 protein in ovarian adenoma were 20.0%(2/10),and in ovarian adenocarcinoma were 62.5%(30/48).There was a significant difference between ovarian adenocarcinoma and ovarian adenoma (P<0.05).3.The positive rates of expression of Ki-67 protein in ovarian adenoma were 30.0% (3/10),and in ovarian adenocarcinoma were 68.8%(33/48).There was a significant difference between ovarian adenocarcinoma and ovarian adenoma(P<0.05).4.The positive rates of expression of AnnexinⅡprotein in well and moderately-differentiated,poorly differentiated of ovarian adenocarcinoma were 81.5%(22/27),80.5%(17/21) respectively,no significant difference among them (P>0.05).The positive expression rates of AnnexinⅡprotein on theⅠandⅡstage,ⅢandⅣstage of ovarian adenocarcinoma were 60.0%(9/15) and 90.9%(30/33) respectively,a significant difference between them(P<0.05).The positive rates of expression of AnnexinⅡprotein on theⅠandⅡstage,ⅢandⅣstage of ovarian adenocarcinoma were 60.0%(9/15) and 90.9%(30/33) respectively,a significant difference between them(P<0.05).The positive rate of expression of AnnexinⅡprotein in the group without lymph node metastasis was 58.3%(7/12).While in the group with lymph node metastasis,it was 88.9%(32/36).A significant difference was between them(P<0.05).5.The positive rates of expression of CyclinD1 protein in well and moderately-differentiated,poorly differentiated of serosity ovarian adenocarcinoma were 42.9%(9/21),80.0%(16/20) respectively,a significant difference among them (P<0.05).The positive expression rates of CyclinD1 protein on theⅠandⅡstage,ⅢandⅣstage of serosity ovarian adenocarcinoma were 33.3%(4/12) and 72.4% (21/29) respectively,a significant difference between them(P<0.05).The positive rate of expression of CyclinD1 protein in the group without lymph node metastasis was 22.2%(2/9).While in the group with lymph node metastasis,it was 71.9%(23/32).A significant difference was between them(P<0.05).6.The positive expression rates of Ki-67 protein in well and moderately-differentiated, poorly differentiated of serosity ovarian adenocarcinoma were 52.4%(11/21),85.0% (17/20) respectively,a significant difference among them(P<0.05).The positive expression rates of Ki-67 protein on theⅠandⅡstage,ⅢandⅣstage of serosity ovarian adenocarcinoma were 50.0%(6/12) and 82.8%(24/29) respectively,a significant difference between them(P<0.05).The positive rate of expression of Ki-67 protein in the group without lymph node metastasis was 44.4%(4/9).While in the group with lymph node metastasis,it was 81.3%(26/32).A significant difference was between them(P<0.05).7.There were positive correlations between the positive expression rates of AnnexinⅡprotein and the expression rates of CyclinD1 and Ki-67 protein in ovarian carcinoma(P<0.05).Conclusions1.By detecting the expression of AnnexinⅡ,CyclinD1 and Ki-67 protein in ovarian adenoma and ovarian adenocarcinoma,we found that the rates of the expression of AnnexinⅡ,CyclinD1 and Ki-67 protein in ovarian adenocarcinoma are higher than those in ovarian adenoma,which indicated that the expression of AnnexinⅡ,CyclinD1 and Ki-67 protein may be correlated with the carcinogenesis of ovarian carcinoma.2.There were no correlations between the expression of AnnexinⅡprotein and the grade of pathology.There were positive correlations between the expression of AnnexinⅡprotein and the stage of FIGO,the lymph node metastasis of ovarian adenocarcinoma,which indicated that the activation of AnnexinⅡprotein may promote the development and metastasis of ovarian adenocarcinoma.3.There were positive correlations between the expression of CyclinD1 protein and the grade of pathology,the stage of FIGO,and the lymph node metastasis of serosity ovarian adenocarcinoma.There were positive correlations between the expression of Ki-67 protein and the grade of pathology,the stage of FIGO and the lymph node metastasis of serosity ovarian adenocarcinoma.They indicated that the activation of CyclinD1 and Ki-67 protein may be correlated with the malignancy level and development and metastasis of serosity ovarian carcinoma.4.The expression of AnnexinⅡprotein has positive correlations with the expressions of CyclinD1 and Ki-67 protein,which showed that the activation of AnnexinⅡprotein could promote cell proliferation.AnnexinⅡprotein could induce carcinogenesis,and promote development of ovarian adenocarcinoma with together CyclinD1 and Ki-67 protein.
