Objective: To study the role of the annexin A5in the occurrence anddevelopment of the human laryngeal carcinoma and its clinicalsignificance,we observed expression of annexin A5in human laryngealcarcinoma tissue.Methods: Fifty-three laryngeal carcinoma tissues and twenty-fiveadjacent normal tissues were collected from department of OtolaryngologyHead and Neck Surgery of the affiliated Hospital of North Sichuan MedicalCollege.All samples were divided into three parts,We sent a part forpathological examination,and made another part into paraffin specimens.Weput the third part in the liquid nitrogen tank saved for later use.All cases oflaryngeal cancer were diagnosed as squamous cell carcinoma by pathologicalexamination.Reverse transcription-polymerase chain reaction (RT-PCR) wasapplied to detect the expression of annexin A5mRNA between the laryngealcarcinoma tissues and the adjacent normal tissues on the RNA level.streptavidin-perosidase (SP) immunohistochemistry method was employed toexamine the expression of the protein of annexin A5between the laryngealcarcinoma tissues and the adjacent normal tissues on the protein level.Result: there has significant difference in the expression of ANXA5mRNA between the laryngeal carcinoma tissues and the adjacent normaltissues.The annexin A5mRNA/β-action of the laryngeal carcinoma tissues andthe adjacent normal tissues are0.57987±0.98881and0.45675±0.10801respectively.There is significant difference between the two grey value(P<0.05).The overexpression rate of annexin A5protein was significantly higher in the laryngeal carcinoma tissues (69.81%)than in the adjacentnormal tissues (32.00%).In the laryngeal carcinoma tissues,theoverexpression rate of annexin A5protein in patients of stages Ⅲ-Ⅳ,withlymphatic metastasis were higher than those of stages Ⅰ-Ⅱ,withoutlymphatic metastasis respectively (P<0.05).The overexpression rate ofannexin A5protein in highly differentiated cancer,moderately differentiatedcancer and poorly differentiated cancer was37.04%(10/27),63.16%(12/19)and85.71%(6/7)(P<0.05).In addition,the overexpression rate of annexin A5protein in patient age,gender and clinical classification has no significantdifference (P>0.05).Conclusion:Both annexin A5mRNA and annexin A5protein areabnormally overexpressed in the laryngeal carcinoma tissues.The expressionlevels of annexin A5protein were closely associated with different clinicalstages,pathologic classification and cervical lymph node metastasis oflaryngeal carcinoma and irrelevant with patient age,gender and clinicalclassification.Annexin A5maybe play an important role in the occurrence anddevelopment of the human laryngeal carcinoma.
