Objective:Through investigating the relation between the change of blood serum tumor necrosis factor-α(TNF-α) in rats with different periods fish bile poisoning and the occurrence and progress of acute kidney injury (AKI) after fish bile poisoning. To approach the probalble pathogenesis of AKI after fish bile poisoning.Methods:1. Looked up documents and preliminary experiments to definite the dosage of Ctenopharyngodon idellus bile.2. Prepared for the model of rats poisoned by Ctenopharyngodon idellus bile in different periods.3. Adopted Enzyme-linked immunosorbent assay (ELISA) to detect the level of blood serum TNF-αin 20 rats poisoned with different periods fish bile and in 10 normal control rats, compared the level of blood serum TNF-αbetween rats with fish bile poisoning and normal control rats, compared the level of blood serum TNF-αamong the interclass rats with fish bile poisoning.4.Removed kindey according to the different period from rats, analyzed the relation between the dose of fish bile poisoning and the degree of kidney pathology damage.Results:1. The level of blood serum TNF-αin rats with fish bile poisoning (22.30±5.81) pg/ml is much higher than that in normal control group (16.23±0.32) pg/ml, which has significant statistical significance (P<0.01). Comparing the level of blood serum TNF-αin rats group with 24 hour group (21.33±0.31) pg/ml and 72 hour group (29.33±1.47) pg/ml are much higher than normal control group,which has significant statistical significance (P<0.01), the 72 hour group is higher than 24 hour group, which has significant statistical significance (P<0.01) and the 24 hour group is higher than normal control group,which has significant statistical significance (P<0.01).2. The longer the fish bile poisoning caused by the more severe the degree of poisoning.3.There are positive correlation that TNF-αlevel and kidney pathological damage.Conclusion:1. The success of the modeling of AKI after fish bile poisoning in rats.2. The longer the fish bile poisoning caused by the more severe the degree of poisoning, serum TNF-αincreased the more obvious.3. The pathogenesis' of fish bile poisoning has intimate relation with the blood serum TNF-α, the blood serum TNF-αhas participated in the morbility of AKI after fish bile poisoning, and in the occurrence and advancement of AKI after fish bile poisoning.
