Objective: To study the influence of IL-33 protein expression in kidney tissue of fish bile poisoning rats after h AMSCs transplation. And the mechanism h AMSCs transplantation was explored on treatment of acute renal tubular necrosis(ATN) by fish bill poisoning.Methods: Fish gall bladder poisoning model rats was preperated by gavage administration with fish bile in the early stage of the research. Experiment has been divided into control group(normal SD rats), model group(fish gall bladder poisoning rats), and cell group( in caudal vein transplantation h AMSCs).At the third,7th and 14 th days after h AMSCs transplantation,paraffin-embedded sections of kidney tissue were stained by immunohistochemical staining. The change of IL-33 protein expression was observed under microscope. Integrated optical density(IOD)value was semiquantitatively calculated by image analysis.Results:(1) The level of renal cortex IL-33 protein expression has no change in control and cell group of at the third,7th and 14 th days after h AMSCs transplantation. The levels of renal cortex IL-33 protein expression were higher at the third and 7th days than at the 14 th days. The levels of IL-33 protein expression were higher in model grop at all of times than in control and cell groups(P<0.05).(2) There are a little of IL-33 protein expression on the kidney nearly pith in control rats,and the levels of IL-33 protein expression are not different at these three times(P>0.05). The IOD value of IL-33 expression are greater at the third day than the 7th day, and at the 7th day than the 14 th day(P<0.05) in model group. While in cell group,The levels of IL-33 protein expression is higher at the third day. But the levels of IL-33 protein expression are significantly decreased at the 7th and 14 th day. The levels of IL-33 expression is lowest at the 14 th day(P<0.05).(3) Compared with control group, the IOD value are higher in model rats at all times(P<0.05). The IOD value of IL-33 expression are lower in cell group at all times compared with model group(P<0.05). And the IOD value of IL-33 expression are higher in cell group only at the third day than in control group. While at the 7th and 14 th day,The level of IL-33 expression has reduced to the level of control group(P>0.05).Conclusion: h AMSCs transplantation treatment play a significant protective effect on acute renal tubular injury of induced fish bill poisioning through decreasing the level of IL-33 protein expression in injuried nearly pith renal tissue, and then relie ving inflammatory injury.
