Influence Of Rkabf On Egfr,tgfr Of Ovarian Granulose Cells In Pco Rat

Object:To explore the effects of Restoring Kidney and Activating Blood Formula (RKABF) has on the expression of EGFR and TGFR in Ovarian Granulose Cells of the rats suffering from rats model of polycystic ovary. So, the study is to expound the relevant function mechanism of RKABF from the aspect of PCOS.Methods:We select seventy-nine SD female rats of six-week old who should have the regular estrum of SPF standard.The rats which feeding in the environment that is consistent with SPF-class animal requirements and these random samples are divided into two groups for contrast:the normal group and the model group (the rate is 1:5),and then the PCO Model of the rats are set by hypodermically injecting Letrozole (LE), normal group are set by 1% Carboxymethyl(CMC).We total need 21 days to complete this step.After this the PCOS Model of the rats and the normal group randomly divided into five groups for contrast, i.e.the first group (model group),the second group(RKABF group of high dose), the third froup(RKABF group of average dose),the fourth group(RKABF group of low dose), the fifth group(western medicine group), and the nomal group,a total of six group,then effect them with different medicine:0.9%Nacl,the RKABF decoction, Ethinylestradiol and Cyproterone Acetate Tablets.This process is a continuous work 3 days once a day, the rest a day, for a total work of 21 times.After that, We randomly selected eight rats in each gorup,and we choose one side of the rats used to separed the ovarian granulose cells and check the expression of EGFR and TGFR in Ovarian Granulosa Cells of the rats. Real-time quantitative PCR determination of gene expression values to explore the effect of RKBAF from the aspects of PCOS rats'ovarian granulose cells.Results:Firstly, EGFR and TGFR date which come from the ovarian granulose cells of the PCO rats were detecred in the low expression.; Secondly, RKABF can improve the expression of EGFR and TGFR's date in PCO rats Ovarian Granulosa Cells.Conclusion:RKABF can improve the expression of EGFR and TGFR in PCO rats Ovarian Granulosa Cells.The results suggest the very good effect which RKABF used in clinical treatment of PCOS that might be associated with RKABF can improve the expression of gene receptor in PCOS Ovarian Granulosa Cells.
