Influence Of RKABF On ER & AR Of Ovarian Granulose Cells In PCO Rat

Object:To construct PCO rat model,by exploring the effects of Restoring Kidney and Activating Blood Formula (RKABF) on the expression of ER and AR in Ovarian Granulose Cells of the rats.So,the study is to expound the relevant function mechanism and its significance of RKABF from the aspect of PCOS.Methods:We select seventy-nine SD female rats of six-week-old which should have the regular estrum of SPF standard and divide these random samples into two groups for contraction:the normal group and the model group (the rate is 1:5).Then the normal group are set by 1% Carboxymethyl(CMC).We totally need 21 days to complete this step.In the model with the preparation of letrozole-carboxy methyl cellulose solution fed rat model of polycystic ovarian pathology.After this the PCOS Model of the rats and the normal group randomly divided into five groups for contraction:After the modeling, the model group were randomly divided into:①model control group(12),with saline,(Qd×3d,rest 1d)×21 times;②high dose group of RKABF (13),with a high dose of orally RKABF,(Qd×3d,rest 1d)×21 times;③RKABF prescription dose group (13rats), RKABF Doses stomach, (Qd×3d,rest 1d)×21 times;④RKABF low dose group (13 rats),RKABF low dose gavage,(Qd×3d,rest ld)×21 times;⑤positive control group (13 rats),up to-35 solution for gavage,(Qd×3d,rest 1d)×21 times.After the intervention period,eight rats in each group randomly separated the side of ovarian granulosa cells,real-time quantitative PCR gene expression values measured to observe the RKABF Decoction on rat ovarian granulosa cells of PCO on the expression of ER and AR.Results:①PCO rat model assessment:The regular estrous cycle changes and the rats has been in diestrus,indicating anovulation; under the light microscope,the model group the formation of polycystic ovary-like changes;the serum LH, T levels of model group are significantly higher than the normal group (P<0.01), E2 levels are significantly lower than the normal group (P<0.05).②PCO rat granulosa cells ER, AR expression:in the model ER, AR levels are significantly lower than normal group (P=0.01);after the application of Restoring Kidney and Activating Blood Formula,the high and low dose ER, AR is significantly higher than model control group (P<0.05).Conclusion:Application of the prepared letrozole PCO rat model of PCOS is an ideal model,the reliability is strong. The results suggest the RKABF can significantly increase the expression of ER,AR in PCOS Ovarian Granulosa Cells.
