Objective:Explored micro-image technology used in studying on diseases and syndromes of traditional Chinese medicine and pharmacodynamics.Through observing acetylphenylhydrazine(APH) blood-deficient rat model and limb ischemia-reperfusion injury (LIRI) rat model of microcirculation changes on the dynamic images in order to discuss characteristics of the multiplicity of blood deficiency and complex nature of compound pathological changes in blood deficiency of Si wu tang. Discussed Si wu tang and other traditional ones can enrich the blood and regulate the blood by studying mechanism of microcirculation.Provided a new pharmacodynamic research methods and experimental basis for studying Chinese medicine micro-dialectically and pharmacodynatics of original recipe of Chinese traditional medicine.Paper was divided into three parts:①theoretical research;②experiment study;③integrated discussion.Methods:Used BI-2000 Medical image analysis system as the main method to observe and analyse the affecting indicators of APH rats as well as LIRI model remedyed by Si wu tang. Employed hematology testing and foot microcirculation detection aids,as the evidence.Besides,applicated BI-2000 medical image analysis system,focusing on observing evolution and intervention on mesenteric microcirculation of APH rats and LIRI by Siwutang.Used Laser microcirculation dynamic analyzer JI-200 to observe influence of microcirculation in rat feet caused by Siwutang;Used hematology analyzer F-820 Sysmex to study blood deficienct rats with normal blood indexes change caused by Siwutang;Used automatic hemorheology analyser LBY-N6K to study hemorheology in APH rats caused by Siwutang.Analysis by synthesis is adopted to analyze experimental results and measurement indicators. To explore mechanism of action and rule about APH and LIRI models caused by Si wu tang.Results:①Used micro-image technology to observe blood-deficient models and microcirculation regulated by Si wu tang directly and continuously perceived through the senses.The overall image test data coincided with detecting methods of clinical test which was routinely used.②Si wu tang could change capillary redness of the skin,red blood cell flow pattern of the APH and LIRI rats,and markedly increased blood flow (BFVA,BFVV),improved the vascular network to pay points,improved the abnormal situation of blood vessel diameter(DV,DA),and there was clear dose-response relationship between them.It could also improve the flow of microcirculation in the feet significantly.③Si wu tang could reduce the whole blood viscosity(WBV),the whole blood reduced viscosity (WBRV),aggregation index(RAI),rigidity index (RI),as well as electrophoresis Index(EI),improve the deformation index(RDI) of APH model rats significantly;and Si wu tang LIRI right there with the same blood flow changes APH trends.④Si wu tang could improve the APH and LIRI model rats general symptoms,reducing feet swelling rate significantly. Microcirculation status of the APH and LIRI model rats both had improved.Conclusion:①ynamic micro-imaging technology can be used as an important morphological microcirculation means of detection for its image clarity,data accuracy,easy to handle,etc.And it is worth being studtied furtherly.②Blood deficiency of mesenteric microcirculation in rat showed characteristics which were deficiency of blood-blood stasis-hemorrhage all three could be seen.They were reciprocally cause and effect.LIRI model had serious microcirculation obstacle.③Papers analysed influence about APH and LIRI models caused by Siwutang from multiple angles and indicators,Si wu tang has a new combination with significant benefits which could enrich the blood-invigorate the circulation of blood-Stop bleeding.Microcirculation of APH and LIRI models were improved by Si wu tang.
