Objective:This study documents the Qiwei Baizhu powder can phosphorylate the IκBα, and then degraded by proteasomes releasing NF-κB for translocation to the nucleus in downstream signaling pathway in JAK/STAT. For the purpose of knowing the mechanic of anti-virus and inducing of AVP(antiviral protein) with Qiwei Baizhu powder.Method:1.We chose to evaluate the response of 5-day-old suckling mice to oral administration of Wa strain of human rotaviruses suspension. Then taking the suckling mice diarrhea to confirm the mock-infected mice is success by McELISA.2.The neonate rats is random allocation to mode group(MG),Qiwei Baizhu powder group(BG),ribavirin group(ZG),inhibitor(Genistein) group(YG),Qiwei Baizhu powder group add inhibito(Genistein) group(BYG) and normal group(NG) totally six groups. Each therapy groups of 5-day-old mice were orally inoculated with either PFU(50/μl) 3times/d of Wa strain virus or a control preparation derived from uninfected MA-104 cells administered in an equivalent volume.3. (1) After 5 days drug therapy, observing the protection for suckling mice's intestinal mucous by hematoxylin and eosin;Using the sandwich ELISA to analyse the pre and post IL-8 level in serum and HRV antigen in stool. (2) Each groups of 5-day-old suckling mice are fed with drugs,and then be killed on the time when in 15min,1h,2h,6h,12h,24h,32, 48h. We using the immunohistochemistry to detect the expression of IκBα, NF-κB in intestinal mucous;Using the magnetic beads to separate the CD3+T lymphocyte,and then adopt the RT-PCR to determine the IκBα,NF-κB mRNA.Using the Western Blotting to detect the IκBαand NF-κB in nuclear and cytosolic of iEL CD3+T extracted respectively.Results:1.After 5 days of treatment, the clearance of rotaviral antigen in stool does not show significant difference between BG and ZG, but higher than the YG and BYG(P<0.05).2. The intestinal mucous is improve properly in BG stained by hematoxylin and eosin, comparing to ZG,YG and BYG (P> 0.05).3. The expression of IκBαin intestinal mucous,IκBαmRNA in CD3+T of iEL and IκBαin CD3+T of iEL cytoplasm are increasly in BG comparing to the ZG,YG and BYG (P<0.05),and decreasely after 12h.4. The expression of NF-κB in intestinal mucous,NF-κB mRNA in CD3+T of iEL and NF-κB in CD3+T of iEL nucleus are lower than MG (P<0.05). The accumulation of NF-κB in BG is lower than in ZG when the times are in 1h,24h,32h and 48h(P<0.05). NF-κB of BG is higher than YG and BYG all the time (P<0.05).5. The pre and post serum IL-8 concentrations of BG are lower than ZG after 5 days interference.Conclusion:Qiwei Baizhu powder can alleviate the suckling mice intestinal lesion, meanwhile lower the IL-8 in serum. In addition, Qiwei Baizhu powder can phosphorylate the IκBαin iEL with HRV infected suckling mice in order to contral the inflammation, meanwhile allowing NF-κB to translocate to the nucleus regulateing stimulate gene transcription.
