| Objective: Based on the clear effect of glycosides isoflavones in Qiwei Baizhu Powder on gut microbiota,the effect of Qiwei Baizhu Powder on intestinal glycoside hydrolase activity in mice with dysmicrobial diarrhea was further studied to provide experimental basis for the therapeutic mechanism of Qiwei Baizhu Powder.Methods:(1)After 15 days of Soybean isoflavones intervention in normal mice,food intake and body weight were recorded.16 S r RNA gene sequencing was used to detect small intestine microbiota,gas chromatography-mass spectrometry(GC-MS)was used to detect short-chain fatty acids(SCFAs)in colon contents,and the colorimetric method was used to analyze sucrase,protease,cellulase,and amylase activities in fresh feces.(2)To establish the methods ofβ-D-glucosidase and β-D-glucuronidase activities;To prepare a mouse model of dysbacteria diarrhea by gavage the Cefradine and gentamicin sulfate suspension,and to detect the activities of β-D-glucosidase and β-D-glucuronidase in different intestinal segments of mice by using the intervention treatment of Qiwei Baizhu Powder decoction,The contents of lactate dehydrogenase(LDH),succinate dehydrogenase(SDH),interleukin 17(IL-17)and lipopolysaccharide(LPS)in serum were determined by enzyme-related immunosorbent assay(ELISA),and malondialdehyde(MDA)in liver tissue were determined by TBA microplate assay.Results:(1)Compared to the normal group,the mice in the Soybean isoflavones group had higher food intake but lower body weight growth rate(P > 0.05);The activities of sucrase,cellulase,and amylase in the intestine of the mice were reduced,and protease activity was increased(P > 0.05);The relative abundance of some beneficial bacteria(Psychrobacter,Lactobacillus,Bacteroides)was increased,the relative abundance of related digestive enzymes producers such as Pseudomonas,Clostridium,Actinomyces and Streptococcus was decreased,Psychrobacter was increased,and the species diversity was increased;In addition,acetic acid,valeric acid,isobutyric acid,isovaleric acid,and caproic acid decreased,while butyric acid and propionic acid increased in the mice treated with Soybean isoflavones;Lactobacillus was a key genus in the small intestine in response to Soybean isoflavones metabolism and production of butyric acid and propionic acid.(2)Assays for β-D-glucosidase and β-D-glucuronidase activities were established.β-D-glucosidase and β-D-glucuronidase activities were significantly decreased(P < 0.05 or P < 0.01)and significantly increased(P < 0.05 or P < 0.01)in the small intestinal contents,small intestinal mucosa,and colonic contents of mice with dysbiosis diarrhea.After treatment with Qiwei Baizhu Powder,β-D-glucosidase and β-D-glucuronidase activities in different intestinal segments were increased in the treated group compared with the normal and self-healing groups;MDA,LDH,IL-17,and LPS levels were decreased,and SDH was increased,with a highly significant effect on LPS(P < 0.01).Conclusion:(1)Soybean isoflavones regulate intestinal microecology,control body weight,and affect digestive enzyme activity;Lactobacillus is a critical genus in the small intestine that responds to Soybean isoflavones metabolism and produces butyric acid and propionic acid;(2)antibiotics decreased β-D-glucosidase and β-D-glucuronidase activities in small intestinal contents and mucosa and colonic contents of mice,and increased activities in colonic mucosa;Qiwei Baizhu Powder can regulate the activities of β-D-glucosidase and β-D-glucuronidase in the intestinal tract of mice with diarrhea as well as energy metabolism,oxidative stress,and inflammatory factors in mice,which is beneficial to restore the metabolic ability of mice and improve the intestinal environment of mice to treat diarrhea. |
