Study On The Gene Therapy Mechanism Of Intra Articular Curcumin Solution Injections In Treating Knee Osteoarthritis Of Rabbits Induced By Papain

Objective: To study the gene therapy mechanism of intra articular curcumin solution injections in treating knee osteoarthritis of rabbits induced by papain .curcumin,as the blocker of activation of the IKKβ, play an important role in the NFКb signaling pathways.many in vitro studies have examined the curcumin as the blocker of activation of the IKKβ,but seldom in vivo studies examined it.so this study wants to examined the curcumin as the blocker of activation of the IKKβin the kneejoint of rabbits with osteoarthritis,to reduce the gene expression of cytokines.to retard the apoptosis of the chondrocytes.Method:Use newzealand rabbits as the experimental animals, Treat these rabbits with paipan solutions to make the osteoarthritis module,and then use curcumin solution, sodium hyaluronate, curcumin with sodium hyaluronate ,Sodium Chloride was used in the control group.Using reverse transcription polymerase chain reaction(RT PCR) to measure the expressions ofCL 2,MMP9,COX 2,IKKβ,Integrinβ1,NFКb expressions Using Tunel to measure the chondrocyte apoptosis condition.Results :After all of the treatments for 4 weeks, curcumin group, sodium hyaluronate group, curcumin with sodium hyaluronate group have the statistical significance in the expressions of CL 2,MMP9,COX 2,IKKβ,Integrinβ1,NFКb expressions compared with the osteoarthritis group. curcumin group, sodium hyaluronate group, curcumin with sodium hyaluronate group have the statistical significance in the chondrocyte apoptdtic index compared with the osteoarthritis group. curcumin with sodium hyaluronate group has the statistical significance in the chondrocyte apoptdtic indexcompared with the curcumin group and sodium hyaluronate group.Conclusion : curcumin is a new gene therapy inhibitors of the in the NFКb signaling pathways, which was confirmed in this vivo experiment,In this experiment sodium hyaluronate also was confirmed the effect of treating the osteoarthritis. The basic mechanism of curcumin in the treatment of osteoarthritis inhibit the activation of the IKKβ,reduce the gene expression of cytokines which induced by the NFКb. Release the apoptosis of the chondrocyte.
