Objective:Study the pharmacognostical characteristics and inherent quality of medicinal materials of C.bungei, provide experimental data and scientific evidence for establishing the quality standards.Methods:First,the genomic total DNA which are from four kinds of Bai shouwu, extracted according to the method of molecular biology.Using specific primers for amplifying,cloned,purification and sequenced,through the soft of blast in NCBI,analysis the sequence of rDNA-ITS from different kinds of Bai shou-wu.Second,through the analysis of experimental research about climatic factors of the localities,culture technics,field processed,charecters of crude drugs and inherent qualityjudged by the guildline of GAP.Three,according to the Pharmacopoeia of the People's Republic of China (2005 Edition) the water, ash and extract contents of the medicinal materials of C.bungei were measured. Fourier transform infrared (FTIR) spectroscopy method was used to investigate a new method for the analysis of IR fingerprint spectra of C.bungei.Results:First, the rDNA-ITS sequence of C.bungei were reported for the first time in the worldwide(GU479037 was the registry number of cultivated species; GU198970 was the wild species.),compared with the cultured and wild species,the similarities is 99%,the relationships among the different varieties and the same variety from the Bai shou-wu were investigated through the soft of DNAMAN,provided the molecular biology basis for the identification of different kinds of plants.Second,the primary SOP revision was established.Third,the quality standards of C.bungei requires as the following:water contents should be not more than 10.00%, total ash not more than 4.00%,water-soluble extract not less than 30.00%, alcohol extract not less than 11.00%.The FTIR fingerprinting and the quality standard of C. bungei were initially set up.Conclusions:First, for the first time it used the method of molecular biology to the identification of C. bungei,the method was easy,accuracy and feasible.Second, completed the standardization planting technique of C. bungei for the first time and revised the first SOP draft.Three,it provided the references for medicinal slices,health productions and chinese materia manufactures to formulate the enterprise standard.Four,these research results are the basis for resource protection and sustainable utilization of C.bungei.
