Capparis spinosa L commonly refers to a type of capparis plants. Most Capparis spinosa L distribute in two major areas of China, XinJiang and XiZang. According to the current researches its ingredients are volatile(isothiocyanate, thymol, palmiticacidetc), indole, thioglycosides, flavonoid, sitosterol, glucosinolate and some novel compounds. Capparis spinosa L possesses other activities, such as antimicrobial and antifungal, anti-inflammatory, antihepatotoxic, antioxidant, hypolipiddemic andhypoglycaemic, diuretic, treating gout. In the present study, it were investigated in tissue culture, the analysis of RAPD polymorphism between seed-derived seedling and regenerated plant.These researches provide a significant scientific basis for Capparis spinosa L.The main results obtained from the experiment as follows:1 Rapid proliferation of Capparis spinosa L.(1)It was disinfected with 70% ethanol to 10s, brushing with water 2 times,then treated with 0.1% HgCl2 for 9 min, brushing with water 5 times, the ratio of survival was 86.7%.(2)Through researching the effect of different hormone combination on budding rate and shoot growth, we found that the medium of MS+NAA 0.1 mg/L+6-BA 0.5mg/L was superior hormone combination, which could be used in the first generative cultivation. The average length of plants reached 3.2 cm after 25 days'cultivation.(3)Through researching the effect of different hormone combination subculture culture, we found that the medium of MS+NAA 0.01 mg/L+6-BA 0.5 mg/L was superior hormone combination, which could be used in the first generative cultivation. The average length of plants reached 3.3 cm after 30 days' cultivation.(4)Roots were induced on the medium of MS+0.05 mg/L NAA+0.1 mg/L IB A. The rooting ratio was 73.3%.(5)The better basal material was sand:pearlite(2:1)and the ratio of survival was 40%.2 The experiment showed that the callus come from stem. The optimal medium for callus induction was the MS medium supplied with 0.6 mg/L 6-BA+1.0mg/L 2,4-D+3.0 mg/L NAA. And the callus cultured on the MS medium supplimented with 6-BA 1.0 mg/L+NAA 0.3 mg/L grew rapidly.3 RAPD show that the rapid propagation of regenerated plants and wild seedling, compared to the DNA levelon on acertain extent the percentage variation for 3.46% that also exists between the iregenerated plants, the percentage variation for 1.50%...
