Capparis spinosa L. is a perennial deciduous liana small shrub,and it is a multiple purpose botany species. Which is used in soil and water conservation, sand-fixing and medical value. In this paper,with the purpose of application Capparis spinosa L. was used experiment which is from Italy Sicily were researched in tissue culture technique and Water stress response of seedlings.First, the tissue culture of axillary bud and rapid propagation in the Capparis spinosa L. were studied. The Capparis spinosa L. introduced from Italian Sicily of which the stem segments with axillary buds were taken as the explants;sterilized by ethanol 75% and Hgcl2 0.5%. Then they were cultured on which were added in the 6-BA0.1mg·L-1 +NAA0.02mg·L-1 and 6-BA0.3mg·L-1+NAA0.05 mg·L-1 of MS medium and DKW medium, it is showed the DKW proliferation medium is better. In the experiment,the suitable medium for every stage were discussed by orthogonal experiment. The results showed: In the inducing period, low hormone level was bene fit for bud multiplication, the suitable medium was DKW+ 0.3mg·L-1 6-BA+0.05 mg·L-1NAA, while in the inducing period, dosage of hormone 6-BA and the shoot multiplication were proportioned positively. But if the dosage of 6-BA enlarged or a long time's culture made in high thick of 6-BA , tissue culture seedling would be more,slim and weak, which are not suitable for root induction,and DKW+0.3mg·L-1 6-BA +0.025mg·L-1NAA is the optimum medium for subculture. The recommended medium for rooting stage was1/2MS +0.2mg·L-1IBA+100~200mg·L-1 VB1, in which the rooting percentages were up to 87%. In addition, the pH is 5.8, special illumine-tion for 14h/d. The plantlets survival rate is up to 92% when transplanted on the mixture(peat soil 3: perlite 1).Physiological response of seedling in water stress is gradually arid soil,the test is greenhouse pot test and the normal water supply for contrast.The 3-year-old seedling,annual seedling and 3-month-old seedling which were transplanted from tissue culture seedling were taken as the explants. The difference of physiological response in 3 stages seedlings were researched.The main results show that:First,it is difference between control group and treatment group, such as the leaf relative water content, relative leaf conductivities, chlorophyll content, MDA content, SOD content,POD content and CAT content of control group which change range were small and stable.Second, with the time physiological indices of treatment group were different,3-year-old seedling showed: relative leaf conductivities, MDA content, SOD content,POD content , CAT content and pro content increased,while annual seedling pro content showed increased at first and then decreased,the chlorophyll is decreased .It is complicated in the test physiological of three months old, relative leaf conductivities, MDA content, pro content increased at first and then decreased, but they decreased slowly in the late, and SOD content were showed increased during the water stress.
