"Dong Qin"Xiangju Pian is a kind of medicine for the treatment of rhinitis and sinusitis and with good results.It is produced by Shanxi xiangju pharmaceutical group with Platycarya Strobilacea Sieb.et Zucc.as raw materials on the basis of the theory of traditional Chinese medicine.However, because the company has been using the traditional production process, resulting in the production of finished products throughout the preparations without quality standards, the difficulty of process control and in the absence of continued competitive capability in the face of foreign products.To meet the urgent need for the actual production,we studied the extraction,purification and in vitro antibacterial activity of medicinal active ingredients from Platycarya Strobilacea Sieb.et Zucc. In this paper, an integral part of the study, mainly completed the following results:(1)Refluent ethanol extraction is a suitable route to extract flavonoids from Platycarya Strobilacea Sieb.et Zucc. Obtained the optimum operation conditions, determined by the method of response surface analysis (RSA) on the basis of the single facor tests,are as follows: temperature of 60℃, alcohol volume fraction of 65%, the ratio of solvent and material 9 mL/g, extract 3 times and extraction time of 2h.The extraction rate of total flavonoids could be up to 4.29%.(2)Macroporous resin is a suitable route to purify flavonoids from Platycarya Strobilacea Sieb.et Zucc.Through static adsorption, desorption, dynamic adsorption, desorption experiments,it is shown that H-327B is an ideal Purification resin. Optimization of process conditions are as follows, adsorption process: concentration of sample solution 8.9mg/mL, with a flow rate of 3BV/h. Elution process: use 90% of ethanol and 5BV ethanol as eluant.The purity of flavonoids rise from 10.2% to 62.72% after purification by macroporous resin.(3) After in vitro antibacterial experiments of four kinds of representative bacteria,which are mainly bacteria infection of sinusitis, rhinitis,it showed that flavonoid extracted from Platycarya Strobilacea Sieb.et Zucc.is one of the main active ingredients. It has strong inhibition of Staphylococcus aureus, Escherichia coli, Streptococcus pneumoniae and Pseudomonas aeruginosa, for inhibition zone diameter of 21.5mm, 21.5mm, 18.5 mm and 17.6mm, MIC are 1.25 mg / mL, 1.25mg/mL, 0.313mg/mL and 0.625mg/mL, MBC are 2.5mg/mL, 1.25mg/mL, 0.625mg/mL and 5mg/mL respectively.The results of this study can help the company to improve production technology and product quality, establish product quality standards, and accelerate the development of second-generation products.It has important practical value and theoretical significance.
