Polygonatum filipes Merr. Ex C. Jeffrey et McEwan belongs to Liliaceae system and Polygonatum mLill specie. It is perennial herbaceous plants, produced in Fujian, Jiangsu, Anhui, Guangdong (North part), and its roots are used for medicinal.It combines medical value, edible value, and admirable value as a whole, which has the very high economic value. Polygonatum filipes taste sweet, flat, and have the function of tonifying Qi, nourishing Yin and invigorating the spleen, moistening the lung, which are mainly used in the treatment of body virtual fatigue, shortness of breath, palpitation of phlegm-heat type dry cough, diabetes, etc. This paper used Polygonatum filipes as the experimental raw materials, and carried on the separation and purification process, in view of researching their anti-oxidation Polygonatum filipes’refined flavonoid extraction technology, macro porous adsorption resin purification, aiming to providing scientific basis of the development of them.1.The Polygonatum filipes were used as the materials, and their optimum extraction technology was researched. The ultrasonic extraction method and bath extraction method were also used to extract the flavonoid of Polygonatum filipes, and this two methods were compared. This paper tested the effect of the feed liquid ratio, extraction temperature, extraction time, and ethanol concentration on the extractive ratio of flavonoid, and optimized the extraction technology, at last found out the best extraction technology conditions. The result showed that the ultrasonic assistant extraction method was better than the bath extraction method, the best extracted conditions of Polygonatum filipes’ flavonoids are:70%ethanol concentration,1:15feed liquid ratio,10min ultrasonic time,80w ultrasonic power. Under that condition, the extraction rate of flavonoids was high to6.14%.2.Investigated the influence of four different macroporous resins such as D101, AB-8, NKA-9and S-8on the observation of index, that is D101, AB-8, NKA-9and S-8. The experiment reported here indicated that the AB-8macroporous adsorption resin has the best adsorption performance on Polygonatum filipes’ flavonoid compounds. Through the single factor experiment and orthogonal experiment, we investigated the influence of adsorption time, sample liquid pH value and adsorption temperature on the adsorption effect. This paper discussed the extraction purification process of Polygonatum filipes’ flavonoid compounds, and confirmed the best craft parameter is that sample liquid pH value is6, adsorption temperature is25℃, adsorption time is8h. 3.The spectrophotometry and related kits were used in the antioxidant experiment, and this paper compared the flavonoids’ crude extract liquids and purification’s scavenging activity on O2-· and·OH free radicals and evaluated their total antioxidant capacity of the Polygonatum filipes. The experiment indicated that the flavonoids extract of Polygonatum filipes has certain antioxidant ability. In the experimental range, the flavonoids extracts of Polygonatum filipes have certain scavenging ability on O2-· and· OH free radicals.
