In this paper, coriander were used as raw material. Anthocyanin extracting, purification, antioxidant activity and antimicrobial activity of the coriander flavonoids were studied. This is provided a theoretical basis for the coriander of developing and using for the further.According to single factor experimental results, The optimum parameters of the ultrasonic assisted extraction process of coriander flavonoids were determined by rotatable central composite experimental design and response surface analysis were:ethanol concentration40%, ultrasonic time:38min, ultrasonic temperature:57℃, ratio of material and solvent:1:30mg/mL. So the coriander flavonoids content was14.02mg/g which was close to the predictive value of14.05mg/g.Selecting4kinds of macroporous resin in the purification experiments of the extractive of coriander flavonoids, AB-8macroporous resin for coriander flavonoids compounds had good adsorption and desorption performance. The optimum purification conditions were determined by adsorption and desorption test of static and dynamic:adsorption time:3h, desorption time:1.5h. The concent ration of sample solution was1.0mg/mL, the pH was6.0, adsorption flow rate was2.0mL/min and the maximum extract was360mL, had got the highest absorption ratio; using50mL70%ethanol as the eluting reagent, and the eluting velocity was2.0mL/min.And purity of the coriander flavonoids were up to56.2%after purificating with macroporous resin, increased3.3times than before purification.The pigments were judged for flavonoids by color reaction, and further judgement were flavonoids by TCL, UV and IR.This study with coriander coarse extract and purification of total flavonoids as raw material, the scavenging free radicals in vitro test, comparison with Vc, the determination of flavonoids of coarse extract and purification of reduction force size and remove DPPH-, hydroxyl free radical, the ability of alkyl free radical and superoxide anion. Get the following conclusion:coriander coarse extract and purification of flavonoids of reducing power was enhanced with the increase of concentration, and had a different degree in four kinds of free radicals scavenging effect. When the concentration of1.0mg/mL, purified content hydroxyl free radical scavenging capacity can reach78.23%; Purification of content of alkyl free radical clearance rate could reach48.57%; To the ultra oxygen anion clearance rate was48.26%; When the concentration of1.5mg/mL, Hydroxyl free radical scavenging capacity could reach83.75%, showed the coriander purified flavonoid content to DPPH· and hydroxyl radicals had good removal effect, alkyl free radical and superoxide anion scavenged effect.Coriander purified flavonoid content was studied in several strains of common antibacterial activity, in penicillin sodium, for comparison, get the following conclusion:coriander purification of flavonoids content bacteriostasis of Escherichia coli was most remarkable, and mucor bacteriostatic effect was not significant. Coriander purified flavonoid content of Escherichia coli, bacillus subtilis, mucor and rhizopus bacteria had certain inhibition, when the concentration of sample purification liquid for30mg/mL, the measured inhibitory circle diameter size in the order:Escherichia coli9.7mm, bacillus subtilis8.3mm, mucor7.5mm, rhizopus11.4mm. The minimal inhibitory concentration were as follows:Escherichia coli5mg/mL, bacillus subtilis lOmg/mL, mucor20mg/mL, rhizopus10mg/mL. Coriander purified flavonoid content of tofu, cucumber and apple had certain inhibition effect, and the bean curd and apple’s anti-corrosion effect was better, cucumber anti-corrosion effect was slightly less.
