Halophilic Archaea Zj204 With The Zj206 The 16s Rdna And The Br Gene Core Sequence Analysis

Extreme halophiles, as a kind of microorganism resources, has been paid more attention in recent years. It has offered many new questions for discussion in theory research , such as microbial physiology, heredity and taxology, life sciences and so on, and it has also offered new materials for life origin and evolution. The photon-driven proton translocator bacteriorhodopsin is considered to be the best understood membrane protein so far. It is nowadays regarded as a model system for photosynthesis, ion pumps and seven transmembrane receptors.Halophilic Archaea AJ6 was located and purified from the Altun Mountain of Xinjiang Uygur Autonomous Region. Strain AJ6 is Gram-negative rods, a few are globular, with the size of single cell of 0.2-0.6 by 1.6-4.2 u m. The optimum salt concentration for its growth is 20% NaCl and 0.6% Mg2+, and the optimum pH is 6-7. Halophilic Archaea ZJ204, ZJ206 was located and purified from the sea water of Sisu salt field of Zhejiang Province. Strain ZJ204, ZJ206 is Gram-positive, rods.Based on its morphology and physiological and biochemical features, 16S rRNA encoding gene (16S rDNA) from AJ6, ZJ204 and ZJ206 were amplified by PCR, and their nucleotide sequences were determined. "MEGA2" and "DNASTAR" software bags were used to analyze 16S rDNA sequence, and the homology was compared and phylogenetic analysis were experted, at the same time, phylogenetic tree of halophilic archaea was established. It was considered that strain AJ6 is a novel species of the genus Natrinema, ZJ204 and ZJ206 are the novel species of the genus Haloferax. The GenBank accession numbers of the 16S rDNA sequences about strain AJ6, ZJ204 and ZJ206 are AY277584, AY838279 and AY838278.In addition, bop gene from ZJ204 and ZJ206 were amplified by PCR, and translated into amino acid sequences. It was found that the differences of the sequences are much in evidence, so it is deserved investigating further. The GenBank accession numbers of the bop gene sequences about strain ZJ204 and ZJ206 are AY838280 and AY838281.
