Yiwu Lake, Xinjiang Archaea Separation And Related Strains Of Bop Gene Core Sequence Analysis

Extreme halophiles, as a new microorganism resources with great application potential, has attracted more and more attentions. They have special physiological conformation and metabolic mechanism, also they can produce a lot of bioactive materials with special properties , which makes the studies on them be valuable both on theory and application. The photon-driven proton translocator bacteriorhodopsin is the only protein in the purple membrane, which is considered to be the best understood membrane proteins so far.There are a lot of salines in Xinjiang. Yiwu lake is one of the inland saline. Because of the special location and entironment status the ecosystem in the lake region is kept well. The salt concentration is as high as 269.91 g/L , which creates a well habitat for halophiles.81 strains were isolated from 15 water samples collected form Yiwu lake. 17 strains were isolated with medium HM, which is used to isolate moderate halophiles and microorganisms which can tolerate high salt concentration. They are brown, yellow ,white or achromatism. 64 strains were isolated with medium B, C, D and E which are used to isolate extreme halophiles. They are red or with similarity colour. Some strains are a little amaranthine, which caused by high density of purple membrane. The red strains are mostly Gram-negative and rod, a few are globular and there are a little strains with special configuration.In addition, bop genes were amplified by PCR from eight red halophilic archaeas isolated form Yiwu lake and four strains from other salines in Xingjing. It is found that there are variety bop genes in the strains from Yiwu lake, which composed two ramifies. The bop genes from Xingjiang has special characteristics, and they are separated in AR,CR and DR, which are the three kinds of BR. It is also found that the differences of the sequences are in evidence, so it is deserved investigating further.
