Extraction, separation and purification of flavonoids compounds from Lilium pumilum bulb, its antioxidant capacity and inhibiting effects is explored, Provide a reference for the development and utilization of Lilium Pumilum. The results as follows:1. The gauging wavelength of rutin after the color was determined by UV-VIS spectrophotometric method. Results of qualitative analysis indicated that the extracts from Lilium pumilum bulb has flavonoids. The method of response surface analysis was adopted to optimize the extraction conditions of total flavonoids form Lilium pumilum bulb on the basis of single-factor experiment. The results showed that the optimum technological conditions were: 73% ethanol concentration, 32:1 m L/g material-liquid ratio, 69℃ reflux temperature and 120 min reflux time. Under such conditions, the actual value of total flavonoids extracted was 4.685 mg/g, which was basically coincident with the predicted value 4.729 mg/g. RSD is 0.661%, the results suggested that the optimum process conditions generated are reliable.2. In order to study the purification effect of AB-8 macroporous resin for total flavonoids from Lilium pumilum bulb, the static and dynamic absorption desorption methods were used, and the content of total flavonoids in samples was determined with UV spectrophotometer. The results showed the optimum process for the adsorption was the crude extract was adjusted to a concentration of 3.165mg/m L, at p H 4, loading at a flow rate of 1.0 m L/min, the flowing of 70% ethanol aqueous solution through AB-8 column was found optimum for the desorption. Under these conditions, the total flavonoid content of extract was increased to 35.13%. AB-8 macroporous adsorption resin was fit for purifying of total flavonoids from Lilium pumilum bulb.3. Three antioxidant assays in vitro(super oxygen anion, hydroxyl free radical scavenging capacity and DPPH radical scavenging capacity) were carried out with the total flavonoids from Lilium pumilum bulb in this study. The results showed that the total flavonoids from Lilium pumilum bulb exhibited strong antioxidant activities, which correlated positively with total flavonoids content. The scavenging rate of total flavonoids from Lilium pumilum bulb on superoxide anion was 63.27%, the scavenging rate of hydroxyl free radical was 70.36% and the scavenging rate of DPPH free radical was 52.83%. those even better than BHT scavenging capacity in the experimental concentration range, the results suggested that the total flavonoids from Lilium pumilum bulb to the selected free radical has a good cleaning effect.4. The bacteriostatic experiment can test the abilities of bacteriostatic of total flavonoids from Lilium pumilum bulb. the minimum inhibitory concentration(MIC) values were: Candida albicans was 80mg/ml, Staphylococcus albuswas 10mg/ml, Shigella flexneri was 80mg/ml, Staphylococcus Lemon was 40mg/m L, Salmonella typhi was 100mg/m L, Rice blast fungus was 60mg/m L, Botrytis cinerea was 80mg/ml, Valsa mali was 40mg/m L, Fusarium oxysporum f. sp. Niveum was 60mg/m L. The total flavonoids from Lilium pumilum bulb on nine kinds of bacteria have different degrees of inhibition, prove it has anti-bacterial properties.
