Study on Pugionium cornutum Leaves as raw material.Extraction, separation and purification of flavonoids compounds in Pugionium cornutum Leaves, explore its antioxidant capacity to use and development Pugionium cornutum resource in the future. The results as follows:(1) Using the response surface design software:ultrasonic assisted and reflux extraction method is used respectively to extract Total Flavonoids from Pugionium cornutum Leaves all-around,through single factor and response surface test, optimize the extraction conditions of Total Flavonoids from Pugionium cornutum Leaves.Specific results are as follows:①By using the response surface method through reflux extraction process to optimize Total Flavonoids from Pugionium cornutum Leaves. The optimum conditions was 85% ethanol concentration, 1:28 ml·g-1liquid ratio, 85℃extraction temperature, 2h reflux time. In this condition the yield of flavonoids is 26.5132mg?g-1 with the theoretical value of RSD was 2.3%. ②By using the response surface method through ultrasonic extraction process to optimize Total Flavonoids from Pugionium cornutum Leaves. The optimum conditions: 80% ethanol concentration, liquid ratio was 1:26 ml·g-1, ultrasonic extraction temperature was 70℃, 42 min ultrasonic time.In this condition: repeated three times the yield of flavonoids is 26.0389mg/g、26.0778mg/g、26.0645mg/g,average 26.0604 mg/g, compared with the theoretical value of 26.0899 mg/g, RSD was 2.086%. Showing the optimal process parameters are accurate and reliable, predicting the analysis of experimental results.Considering the subsequent big workload,short time,the follow-up work select ultrasonic extraction method in the following-up work.(2) Using orthogonal design method to optimize the AB 8 macroporous resin purification of total Flavonoids from Pugionium cornutum Leaves, the optimal process parameters are as follows:The optimum adsorption conditions: the sample’s solution concentration was 1.96 mg/m L, the sample solution’s feeding speed was 2.0 m L/min, p H 5 sample liquid, maximum sample amount 5 bv, verified the adsorption rate was 87.62%.The optimum conditions: 80% ethanol, 2.0 m L/min elution speed, 4BV elution volume, verified the amount of parsing is 1.84 mg/g.The recovery was 83.16%, the purity from 22.16% to 64.93%, refined multiples 2.93 by measuring, showing the AB 8 macroporous adsorption resin to Total Flavonoids from Pugionium cornutum Leaves has better purification effect of refined, the resin has good selectivity, high resolution、adsorption, operation is simple and harmless.(3) Four kinds of in vitro antioxidant system results show that Pugionium cornutum Leaves’ crude extract and purification of total flavonoids content for Fe3+ having a strong ability of reduction, also having a clearance ability for ·OH, O2-, DPPH, through comparing the overall antioxidant capacity size order: purified content > butylated hydroxytoluene > crude extract.
