Ramie is a perennial herb branch in urticaceae and boehmeria, which is an economically important crop.It provides fibers used for textiles.The some4%of entire plants is usefull,the rest of entire plants are abandoned as by-products.The modem medical research,flavonoids have various kinds of pharmacological activities,this paper studies on extraction and purification condition of flavonoids and its contents differences in genotype in ramie,result:1It studies by means of single factor experiment and orthogonal experiment and validation test methods, the optimum extraction condition of alcohol reflux extraction is that the material liquid ratio is1:70and the concentration of ethanol is80%and the extraction time is100minutes and the temperature is80℃,here the average extraction rate of total flavonoids in ramie is3.75%.The optimum extraction condition of extraction of cellulase is that the cellulasa dosage is3mg and enzymatic hydrolysis pH=4.5and the enzymatic hydrolasis time is80minutes and the temperature is50℃, here the average extraction rate of total flavonoids in ramie is3.82%.The optimum extraction condition of ultrasonic assisted extraction is that the material liquid ratio is1:35and ultrasonic power is100W and the ultrasonic time is45minutes and the temperature is60℃, here the average extraction rate of total flavonoids in ramie is4.21%.2By comparing the performance of adsorption and desorption in3kinds of resins:AB-8S-8, and D4020, the AB-8type resin was determined to to purify flavonoids in ramie. The best optimization adsorption and desorption parameters of AB-8type resin has been found by static adsorption and desorption experiments of AB-8type resin,dynamic adsorption and desorption experiments of AB-8type resin and single factor experiment and orthogonal experiment. The optimum adsorption condition is that the sample concentration is0.3mg/ml and the sample flow is0.2ml/min and the sample pH=6. The optimum desorption condition is that the eluate concentration is95%and the eluate flow is3ml/min and the eluate dosage is5BV.3The study found that the variation range of the content of total flavonoids in ramie is0.209-4.748%and the average of total falvonoids is1.42%, when the dynamics analysis of total flavonoids in different parts and sampling time in20kinds of ramie was researched. The highest content of leaf’s total flavonoids in ramie variety is "Chengbu local ramie", of which the content is4.748%, The lowest content of leaf’s total flavonoids in ramie variety is "Leiyang tiesi ramie", of which the content is0.336%. The content of total flavonoids in diverse plant parts are different, the descending order of the content of total flavonoids is lesf, root and stem,of which the aveange content were determined to be2.309%,0.689%and0.643%. The contents of total flavoniods are increasing in the period of ramie growth and growing process.The average content of total flavonoids have reached the highest levels,which the content is2.702%,when ramie is in late colsure line. The average content of total flavonoids have reached the lowest levels, which the content is1.91%before the closure stage.The20varieties of ramie are divided into3types.There are3varieties of high total flavonoids ramie,10varieties of middle total flavonoids ramie and7varieties of low total flavonoids ramie.
