A systematic study had been conducted on the polyphasic taxonomy of TRM49590with the polyphasic taxonomy to determine its and the strain TRM49590was isolated from soil samples lop nur in xinjiang. In order to obtain bioactive secondary metabolites, the second metabolites of TRM49590were separation and purification with the tracking of antibacterial activity.1. The polyphasic taxonomy of TRM49590A novel Streptomyces strain, designated strain TRM49590T was isolated from Lop Nur in Xinjiang, China. Strain TRM49590T was aerobic, Gram staining positive, with an optimum pH for growth of pH9, an optimum NaCl concentration for growth of1.5%(w/v) and an optimum temperature for growth of28-37℃. The aerial mycelia were sparse and cylindrical, with irregular branches and smooth-surfaced on ISP medium4. The whole-cell sugar of strain TRM49590T was ribose and glucose. The diagnostic diamino acid contained LL-diaminopimelic acid. The predominant menaquinones were MK-9(H4), MK-9(H6), MK-9(H8) and MK-10(H6). The major fatty acids were iso-C16:0, anteiso-Ci5:0and anteiso-C17:0. The G+C content of the genomic DNA was62.2mol%. The major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, phosphatidylinositol and phosphatidylinositol mannoside. Phylogenetic analysis based on16S rRNA gene sequence analysis showed that strain TRM49590T belong to the genus Streptomyces with sequence similarity of97.16%with the closest described species Streptomyces sodiiphilus. Based on these observations and other physiological features, the strain TRM49590T is proposed to represent a novel species of the genus Streptomyces for which the name Streptomyces lopnurensis sp. nov. is proposed. The type strain is TRM49590T (=CCTCC AA2013018T=NRRL B591091).2. The research on secondary metabolites of Streptomyces lopnurensis TRM49590The Streptomyces lopnurensis TRM49590was selected for the research。We carried out methods for screening the antibiotic functional genes with the molecular biology method, separating and purificating the compounds from the fermented liquid and bacteria extraction with the tracking of antibacterial activity, such as macroporous resin, silica column chromatography, gel column chromatography, high performance liquid chromatography (HPLC), and the preparation of liquid chromatography etc. We carried out methods for identifying the compounds, such as nuclear magnetic resonance (NMR) etc. The results showed that Streptomyces lopnurensis TRM49590contained1kind of antibiotic functional genes, which is HMG-CoA, The fermentation of Streptomyces lopnurensis TRM49590has the antibacterial activity to Fusarium oxysporum f. sp. vasinfectum ATCC31083, Verticillium dahliae ATCC36211, Staphylococcus aureus ATCC25923and Staphylococcus epidermidis ATCC35984. The component A has the antibacterial activity to Fusarium oxysporum f. sp. vasinfectum ATCC31083and Verticillium dahliae ATCC36211, The component D has the antibacterial activity to Staphylococcus aureus ATCC25923and Staphylococcus epidermidis ATCC35984. The bacteria extraction has the antibacterial activity to Fusarium oxysporum f. sp. vasinfectum ATCC31083. Eventually we isolated1compound from the fermented liquid, which is N-acetyltyramine.
