Two potential new species of TRM 46515 and TRM 46509 from Alar,Xinjiang province were subjected to multiphase taxonomy investigation.The secondary metabolites from new species of TRM46515 were isolated and identified via chroma to graphy and NMR methods.Multiple screening methods inclonding PCR of biosynthetic enzyme gene/bioassay/HPLC-DAD test were established in order searching for actinomycetes being able to produce polyene macrolide antibiotics.The polyphasic taxonomy of new Streptomyces of TRM 46515 and TRM 46509 were studied.The polyphasic taxonomy of two new species of actinomycetes was carried out on the basis of their morphologicalcharacteristics,phylogeneticcharacteristics,physiologicaland biochemical characteristics and cytochemical characteristics.The results showed that the actinomycetes TRM46515 and TRM 46509 were new species of Streptomyces and identified as Streptomyces litoralis and Streptomyces kalpinensis,respectively.Fermentation broth of Streptomyces litoralis TRM 46515 was collected and it metabolites were isolated using macroporous resin enrichment,silica gel column chromatography and HPLC preparative methods.Resulting in obtaining two compounds.Then UV,1H NMR and 13C NMR data analysis together with HSQC and HMBC analysis were used to identified the structure of two compounds as dibutylphthalate and di?2-ethylhexyl?phthalate.Method of designing?via co-linear analysis?and PCR of KR,DH and TE genes,combining with the polyene macrolide antibiotic characteristics analysis of ultraviolet absorption and antibacterial activity test against Candida albicans were established.The methods were proved to from cotton field screened being cable of seaching for PMA from strain 4.1116.Furthermore,one strain TRM 20904was found out to detecting KR,DH and TE genes as well as showing antifungal activity against C.albicans and UV?max=362,382,404nm.
