Prunus is a kind of economic macadamia trees (including Prunus salicinaLindl., Prunus armeniaca), after a long-term evolution and breeding, there were rich varieties. The study on the genetic diversity of almond benefits the collection, conservation, identification and reasonable use of the the germplasm.In this study, the RAPD makers were used to detect the genetic diversity and cluser analysis on20prunus materials and established the DNA fingerprints. A new strategy named CID was first employed for identification of Prunus materials. From the location of these cultivars in the CID, it was easy to find the primers to be used to separate them to further research. Confirming CID strategy was definitely feasible, efficient, referable and practicable and will be very useful for the protection of cultivar rights in the nursery industry.The main results were as follows:1.This study adopt the SDS method, CTAB method, one-step method three methods of DNA extraction, extracting prunus material of blade and fresh buds, the method about extraction and purification of almond genetic DNA was set.2.Using the research results of other nut plants, the RAPD amplification programs were optimized to set up the reaction system of RAPD fitting for analyzing the resources of almond with the orthogonal diagram.3.Respectively from20RAPD primers (10-mer), picking out the suitable six RAPD random primers with polymorphic and informative patterns for analysis. Six of random primers for analysis of RAPD products were amplified from234strips,208polymorphic bands were produced, the percentage of polymorphic bands was88.90%. Every primer can generate1to7amplified bands. According to the amplified bands of0,1matrix, using UPGMA method to cluster analysis of20test materials. The results showed that:RAPD markers to reveal the genetic distance between prunus germplasm materials range in0.05to0.50, with a mean of0.28. All these20prunus materials could be clustered into6groups by UPGMA method.4.By amplification of primer Y18(11-mer), initially built a CID fingerprint of20prunus materials.
