The ACC oxidase and synthase genes were cloned from genomic DNA of Prunus persica Batsch and Prunus pseudocerasus L. by analyzing DNA homogeneous and designing directional primer.PCR amplified products were collected, Conneeted and transformed after they were examined by 1 % agarose gel electrophoresis. The white clony was selected and the plasmid DNA was isolated and identified by restriction enzyme analysis, plasmid PCR. After recombinant plasmid was identified, the DNA sequence was determined. The suitable ACC oxidase and ACC synthase gene cloning PCR reacting system,exogenous DNA fragment connection reacting system and recombinant plasmid restriction enzyme analysis identfication reating system were selected. The results of DNA sequencing revealed that ACC oxidase gene sequence of Prunus persica is 1313bp and ACC oxidase gene sequence of Prunus pseudocerasus is 131 Obp. They all included 2 exons and 3 introns and the full length of exons was l000bp. The ACC oxidase gene mRNA sequence, duced amino acid of Prunus persica and Prunus armeniaca were 94.1% and 96.7% homologous respectively, and ACC oxidase gene DNA sequence of Prunus pseudocerasus and Prunus persica were 97.8% homologous.In addition, ACC synthase gene sequence length of Prunus persica was 1416bp, ACC synthase gene mRNA sequence of Prunus salicina were 71.2% homologous.
