The novel detection technology of Surface Plasmon Resonance (SPR) assay is known to monitor interaction between bimolecular real time and ascertain the kinds of reactants as well as their concentrate without isolation and purification. SPR sensors are valuable analytical tools with abroad range of applications from laboratory assays to study for food safety, drug discovery and environmental pollutant monitoring, which have the feature of high sensitivity, label free, small amount of sample and rapid speed of detection. Study and development for a compact, inexpensive SPR detection instrument is very important.Based on the numerous researches works and analyzing for the background and principle of SPR technology and its developing trend. This paper introduces the development and experimental application of a new SPR detection instrument.The paper analyzes the principle of SPR and designs the structure of SPR detection system, develops a angle sweeping system which can precisely change the incident angle of light source from 45°to 72°.The paper designs and manufactures the hardware of SPR detection system, including the electro-optic switching circuit, the signal amplifying circuit, the A/D conversion circuit, the control circuit and the USB communication circuit. The paper designs and develops the software of SPR detection system, including the firmware program which controls the hardware and completes the detection, the user application program with friendly interface to process and store data, and the USB driver.The paper analyzes and determines stability of the angle sweeping system and resolution of the SPR detection system. Results show that the system is stable and it can reach a high resolution.
