Purpose:This study based on the hyperplasia of mammary glands in rats with treatment of EA “Tanzhong”(CV17), “Zusanli”(ST36)and “Taichong”(LR3)point to observe the expression of Bcl-2, P16 and P53 content in breast tissue, discusse whether abnormal apoptosis is one of the pathological factors of hyperplasia of mammary glands and confirme the effect and mechanism of electroacupuncture treatment on hyperplasia of mammary glands.Materials and methods:40 SPF SD unfertilized female rats were randomly divided into 4 groups, each group of 10 rats, respectively, for the normal group, model group, acupuncture group, and tamoxifen group. After the success of the modeling, each group was filled with ordinary feed. Model group were only fixed on the table 15 min /d without other treatment; According to the<<experimental acupuncture>>, acupuncture group were selected the “Danzhong”, bilateral “Zusanli” and bilateral“Taichong” point needling into about 3 mm with electroacupuncture treatment(Electroacupuncture Parameters: density wave)for 15min/d and strength of the needle handle trembling; Tamoxifen group were administered with water solution 1.8 mg·kg-1/d by gastrogavage for 4 successive weeks.After the treatment, the breast tissue of each group was taken down, and the morphology of the nipple was observed by measuring the diameter of the nipple and HE staining. The expression of Bcl-2, P16 and P53 were detected by real-time fluorescence quantitative PCR method. The protein expression of P16 and P53 were detected by immunohistochemical method and the light density was calculated.Results:1. The influence of electroacupuncture “Tanzhong”, bilateral “Zusanli”, bilateral “Taichong” on the nipple diameter of each group.Compared with the normal group, the second and third paired nipples were was significantly greater in model group(P<0.01), nipples in acupuncture group and tamoxifen group had no obvious change(P>0.05); Compared with the model group, nipples was significantly smaller in acupuncture group and tamoxifen group(P< 0.01);2. Observation of the morphology of each groupIn the normal group, the number of mammary glands is less, the size of the mammary gland was uniform, the arrangement was regular, there was no lymphocyte infiltration and no secretions or less in the alveolar cavity, and no extension of the duct; In the model group, glands of mammary gland were hyperplastic, mammary gland flocculus significantly increased, gland bubble expanded and had papillary changes, and a large amount of secretion in the acinus cavity and duct cavity; In the acupuncture group and tamoxifen group, expansion of gland and duct was significantly reduceed, numbers of mammary gland flocculus were significantly decreased, mammary ductal epithelial hyperplasia was weakened, and secretion declined significantly.3. The influence of electroacupuncture “Tanzhong”, bilateral “Zusanli”, bilateral “Taichong” on the Bcl-2 content in mammary tissue of each group.Compared with the normal group, Bcl-2 in breast tissue of model rats were significantly increased(P < 0.01); Compared with the model group, Bcl-2 in breast tissue decreased significantly in acupuncture group and tamoxifen group(P < 0.01).4. The influence of electroacupuncture “Tanzhong”, bilateral “Zusanli”, bilateral “Taichong” on the P16 content in mammary tissue of each group.Compared with the normal group, P16 in breast tissue of model rats were significantly increased(P < 0.01); Compared with the model group, P16 in breast tissue decreased significantly in acupuncture group and tamoxifen group(P < 0.01).5. The influence of electroacupuncture “Tanzhong”, bilateral “Zusanli”, bilateral “Taichong” on the P53 content in mammary tissue of each group.Compared with the normal group, P53 in breast tissue of model rats were significantly increased(P < 0.01); Compared with the model group, P53 in breast tissue decreased significantly in acupuncture group and tamoxifen group(P < 0.01).Conclusion:1. Electroacupuncture “Tanzhong”, bilateral “Zusanli” and bilateral “Taichong” could effectively improve nipple diameter and pathological structure of the breast tissue in mammary glands in rats.2. There were abnormal expression of apoptosis factors in mammary gland hyperplasia rats, and it may be one of the pathogenesis of mammary gland hyperplasia.3. Electroacupuncture “Tanzhong”, bilateral “Zusanli” and bilateral “Taichong” may control the expression of apoptosis factors to protect mammary gland tissue of mammary gland hyperplasia rats against apoptosis and reverse the hyperplasia of breast tissue.
