ObjectiveBy cutting off the vagus nerve as intervention,to study whether the effect of electric acupuncture treatment of mammary hyperplasia mechanism is related with the vagus nerve.MethodsTo select 62 healthy,unfertilized female SD rats(180-220 g),and randomly divided them into 5 groups:Blank group(group A),model group(group B),the treatment group(group C),the vagus nerve resection and treatment group(group D)and sham operation(group E).Blank group and model group each had 13 rats,and the other three groups each had 12 rats.Rats in the blank group were injected with 0.9%saline,0.1 ml,1 time/day for 30 consecutive days;Rats in the other four groups were given intramuscular injection estradiol benzoate at a dose of 0.5 mg/kg/d for 25 consecutive days,followed by intramuscular injection of progesterone at a dose of 0.5 mg/kg/d for5 consecutive days.On the basis of this model,the vagus nerve treatment group took the right side of the neck vagotomy as the intervention method,and the sham operation group only expose but not to resect the vagus nerve.Except the blank group and the model group,the remaining three groups were treaten with electroacupuncture.The frequency of electroacupuncture was 2Hz,the waveform was the dense wave,the treatment time was 5min,once a day and 5times as a course of treatment,a total of four courses of treatment.2 days of rest during each course of treatment,then the next course of treatment will be continued.The experiment will be carried out with the following tests:1.To measure rat's nipple height in various stages such as before modeling,after modeling,and after treatment;2.HE conventional staining,to do breast histomorphology observation and analysis under the light microscope;3.taking flow cytometry tests the percentage of lymphocyte subsets in rats'peripheral blood.Results1.After modeling,the nipples'height in group B,group C,group D and group E significantly exceeded that in group A(P<0.01).After treatment,the height of group C was significantly lower than that of group B(P<0.01)There was no significant difference between group D and group B(P>0.05).The height of nipple in group E was significantly lower than that in group D(P<0.01).2.Pathological findings of breast tissue:The ductal epithelium of the breast tissue of group A was arranged in order,and there was no expansion in the duct and acinar cavity.The ductal epithelium of the group B was in disorder,the degree of glandular epithelial hyperplasia increased,and there was more secretion in the duct lumen and acinar Compared with group B,the ductal epithelium of group C was basically regular and the acinar dilatation was not obvious.In group D,the ductal epithelium of breast tissue wasirregular and the number of acinar cells was significantly higher than that of group B More,breast duct and acinar is still expanding;E group of breast ductal epithelial cells arranged in the basic rules,and only a little of hyperplasia in breast lobular and acinar hyperplasia,and slight degreedilation in breast duct.3.The percentage of T lymphocytes in each group was compared:Compared with group A,the percentage of CD8~+in group B was significantly lower than that in group B(P<0.01),and the percentage of CD4~+/CD8~+in group B was significantly higher than that in group B(P<0.01).Compared with group B,the ratio of CD8~+and CD4~+/CD8~+in group D was not significantly different from that in group B(P>0.05).Compared with group D,the percentage of CD8~+in group D was significantly higher than that in group B(P<0.01),CD4~+/CD8~+ratio was significantly lower than that of group D(P<0.01).Conclusion1.The mechanism of electroacupuncture on mammary gland hyperplasia is related to vagus nerve.2.The regulation of Electroacupuncture on T-lymphocyte subsets in mammary gland hyperplasia is related to vagus nerve.
