Effect Of Acupuncture Combined With Qiqi Qufeng Decoction On Serum IFN - γ And IL - 4 Contents In ACD Model Mice

Purpose: This experiment aimed to detect the imbalance of ACD model rat serum Th1/Th2 by acupuncturing Xuehai, Quchi, Tianjing combined with Yiqiqufeng decoction. To put forward a preliminary discussion of immunological mechanism on ACD acupuncture treatment. To provide the theoretical basis for the treatment of ACD with the combination of acupuncture and decoction in clinic.Material and method:The 60 healthy male Kunming mice were divided into 6 groups by random grouping method, respectively contract group, ACD model group, positive drug control group, Yiqiqufeng decoction group, acupuncture group, acupuncture combined with decoction group. Use DNFB to induce the ACD in mice model. All the drugs were administered by gastric perfusion method, of which the positive drug control group with Loratadine tablets coagulation liquid dissolved by distilled water. Acupuncture and acupuncture combined with decoction groups both be treated with Yiqiqufeng granules. Whereas, the control group, model group and acupuncture group be treated with distilled water meanwhile. To observe the treatment of acupuncture combined with decoction on the influence and lesion morphology of ACD mouse ear swelling lesions. Adopt ELISA method to detect the IFN-γ,IL-4 content, in order to explore the immunological mechanism of acupuncture and decoction treatment on ACD from the point of the balance of TH1/TH2 cytokine network.Results:1. Acupuncture combined with decoction treatment can down regulate the weight difference and swelling degree of the sick ear of the mice.The swelling degree of the sick ears of the model group mice is remarkably higher than the control group(p<0.01). And the swelling degree of the sick ears of the acupuncture combined with decoction group, acupuncture group, decoction group, positive drug control group is remarkably lower than the model group(p<0.01 or p<0.05).2. The effect of acupuncture combined with decoction treatment on IFN-γand IL-4 in ACD model mice serum.The IFN-γin serum of ACD model group mice is remarkably higher than the control group(p<0.01), however the IL-4 is lower than that of the control group(p<0.01). The IFN-γin serum of acupuncture combined with decoction group, acupuncture group, decoction group and positive drug control group is all less than that in model group(p<0.01). Whereas, the IL-4 is higher than that of the ACD model group(p<0.01).Conclusion:1.Acupuncture combined with Yiqi Qufeng Decoction has a specific therapeutical effect for ACD mouse model, and can reduce swelling degree of the diseased pinna.2.The levels of IFN-γ raised, IL-4 reduced, Th1/Th2 unbalanced in ACD mouse mode serum.3.Acupuncture combined with Yiqi Qufeng Decoction can reduce the levels of IFN-γ,increase the levels of IL-4, correct Th1/Th2 unbalance of ACD mouse mode serum,and may be one of the mechanisms for treat the ACD.
