Effect Of Acupuncture At "Feishu" Point On IL-4, IL-6, IFN-γ And ACTH In Rats With Lung Immune Inflammation

Purpose:This experiment by inducing Ovalbumin to establish the rat model with bronchial asthma,and acupuncturing bronchial asthma rat’s “Feishu” points, observing the symptom of asthma,and the expression of rat’s alveolar lavage fluid and serum IL- 4,IL- 6, IFN- gamma and IL- 6 in the lung tissue and ACTH contents in serum, evaluating,by acupuncture stimulating Feishu points,the influence of the mediation of the nervous immune endocrine system on inflammatory response in rats with bronchial asthma,and further investigating the intervention effect of IL-4,IL-6,IFN-γ,and ACTH,by acupuncture stimulating "Feishu" points.Materials and methods:According to the table of random numbers, the only 200 g of 40 male Wistar rats divided randomly into four groups(10 in each group) and normal contrast group(0, 7, 14, 21 days, abdominal injection of PBS, 22 days to 43 days 1% Na Cl atomization inhalation,20 minutes each time);model group(0, 7, 14, 21 days, intraperitoneal injection of contain OVA1 mg + aluminum hydroxide(AL(OH) 3) 200 mg of 1 m L of PBS,22 days to 43 days,atomization inhalation of 1% OVA Na Cl solution for 20 minutes, induced asthma); Feishu points group(acupuncturing Feishu 3-5 mm, retaining needle for 20 minutes, moderate intensity, no body twist, every other day at a time and for 21 days), non-meridians and non-acupoint group(acupuncture Feishu and open the shoulder blades about 0.5cm, the retaining needle for 20 min, every other day at a time, and for 21 days). After treatment,collecting the rat’s alveolar lavage fluid, serum and lung tissue, taking use of the method of Elisa to detect the content of IL- 4, IFN – gamma, IL-6 in the alveolar lavage, and to detect the content of IL- 4, IFN- gamma, IL-6 in the serum, and to detect the content of IL-6 in lung tissue,utilizing the method Jim staining in Switzerland to detect the content of alveolar lavage’s inflammatory factor and the content of EOS cells.With the method of HE staining observing the pathological changes of lung tissue.The data used SPSS19.0 statistical software for the analysis of single factor variance, and used the LSD method to analysis the comparison between groups, the results to mean + /-standard deviation(x±s) said, p < 0.05 showed statistically significant difference.Result:1 The changes of lung tissue and tracheal morphologyThe normal group:Epithelial cells arranged neatly and regularly.There was no secretion in the lumens,and the alveolar was normal.Hyperplasia of the lumens and airway smooth muscle did not occur. The asthma group:Epithelial cells arranged irreguarly.The dilatation of alveolar ducts and the edema of the mucous of alveolar ducts were palpable,with the secretion increased. EOS infiltration in bronchial wall occured,and the wall were thickened.The "Feishu" group :Epithelial cells arranged in order slightly,with the mucosa shedding.The lumens and airway smooth muscle were thickened slightly,and EOS infiltration decreased obviously. The non acupoint group:The changes,compared with model groups,were not obvious.2 The total number of inflammatory cells and EOS in BALFCompared with the model group, the total number of inflammatory cells and EOS in "Feishu" group were significantly decreased(P<0.05),and the total number of inflammatory cells and EOS in non acupoint group were significantly decreased(P<0.05).Compared with the normal group,the total number of inflammatory cells and EOS in model group were significantly increased(P < 0.05),and the total number of inflammatory cells and EOS in“Feishu” group were significantly increased(P < 0.05).The total number of inflammatory cells and EOS in non acupoint group were significantly increased(P<0.05).3 The intervention effect of IL-4 with "Feishu" points.Compared with the normal group,the IL-4 contents in model group were significantly increased(P < 0.05).Compared with the model group,the IL-4 contents in “Feishu” group were significantly decreased(P<0.05).Compared with the model group,the IL-4 contents in non acupoint group were not affected(P>0.05).Compared with the “Feishu” group,the IL-4contents of serum in non acupoint group were significantly decreased(P<0.05).4 The intervention effect of IL-6 with "Feishu" points.Compared with the normal group,the IL-6 contents in model group were significantly increased(P<0.05).Compared with the model group,the IL-6 contents in “Feishu” group were significantly decreased(P<0.05).Compared with the model group,the IL-6 contents in non acupoint group were not affected(P>0.05).5 The intervention effect of IFN-γwith "Feishu" points.Compared with the normal group,the IFN-γcontents in model group were significantly decreased(P<0.05).Compared with the model group,the IFN-γcontents in “Feishu” group were significantly increased(P<0.05).Compared with the model group,the IFN-γ contents in non acupoint group were not affected(P > 0.05).Compared with the “Feishu” group,the IFN-γcontents of serum in non acupoint group were significantly decreased(P<0.05).6 The intervention effect of ACTH with "Feishu" points.Compared with the normal group,the ACTH contents in model group were significantly decreased(P<0.05).Compared with the model group,the ACTH contents in “Feishu” group were significantly increased(P<0.05).Compared with the model group,the ACTH contents in non acupoint group were not affected(P>0.05).Conclusion:1 Acupuncture stimulating “Feishu”points can reduce the total number of inflammatory cells and EOS in BALF of lung immune inflammation rats.2 Acupuncture stimulating “Feishu”points can reduce the contents of IL-4 in BALF and serum of rats.3 Acupuncture stimulating “Feishu”points can reduce the contents of IL-6 in BALF,serum and lung of rats.4 Acupuncture stimulating “Feishu”points can increase the contents of IFN-γ in BALF and serum of rats.5 Acupuncture stimulating “Feishu”points can increase the contents of ACTH in serum of rats.
